The mammalian collagen had been widely used in tissue engineering for its goodbiocompatibility and extensive source. But the appearance of mad cow disease andother zoonotic diseases increased its security risks. It is necessary to find the moresecure and biocompatible natural polymer materials. We take aquatic collagen andpolysaccharide of traditional Chinese medicine as the candidate. First we study theextraction, purification, physico-chemical properties and biological activity of threekinds of polysaccharide and squid skin collagen. The results shown that the Chuanxiongpolysaccharide had the best cell growth capacity than other two kinds ofpolysaccharides, and squid skin collagen showed good biocompatibility, absorbent andemulsifying capacity. Then we synthetized Chuanxiong polysaccharide/sqiud collagencomposite scaffold, we found the composite, in the condition ofpolysaccharide-collagen ratio2:8and2.5%glutaraldehyde concentration, showedexcellent biomaterial science features, the tensile strength was0.35MPa; the elongationrate was80%; water absorption was567.12%and aperture volume fration was2500%.These properties indicated that the composite has good mechanical property andwater-wet behavior. Observation by electron microscopy found the porous compositehas almost homogeneous pore size of130μm. In the same procedure, the crosslinkedcomposite scaffold had a long degradation time, about1.5times of the crosslinked purecollagen scaffold. More outstanding is the composite scaffold showed goodbiocompatibility and proliferation promoting activity. Compared with the negativecontrol only with culture medium, the composite scaffold showed more durable andstrong cell growth and proliferation promoting activity, the promoting rate is18%higher than that of pure collagen scaffold. All these properties give Chuanxiongpolysaccharide/sqiud collagen composite good prospects in tissue engineering.
