ObjectiveThe exactly pathogenesis of SLE is not clear. The commonly o-pinion is that the environments, including the results of sex hormone act on body with certain heredity factors. The patient's conditions have related with his genetic background. It has been proved that IL-10 concentration of patients with SLE producing by monocyte in peripheral blood is much higher than that of normal person, and has related with the severity level of the disease. The serum level of IL - 10 has shown a correlation with SLEDAI, which can change with the variety of the post - treated IL-10 level. Only when IL-10 binding with its receptor can it play its roles. So we consider that IL-10R may have any correlation with SLE, too. KATHY L. MOSER et al found that there are 14 major opportunit predisposing genes site of SLE. Among them, 1q31, 11q23 is the site of IL-10 and IL - 10R1 code gene respectively. All of these display that IL - 10 and IL - 10R1 have important contact with the pathogenesis of SLE. In our investigation, we find that IL - 10R1 gene disorder in NZW mouse downregulate IL -10R1 on the surface of immunocyte and we also detect IL - 10R1 gene is the predisposing genes of SLE. So, we detect IL - 10R on the surface of T lymphocyte in the patients with SLE in order to explore if the changes of IL - 10R on the surface of T lymphocyte have any relation with SLE and activity of the disease.Methods43 SLE active phase patients were taken as case group, and divided them into treatment and non - treatment groups (20 and 23 respectively) ; 25 non - treatment RA patients were taken as connective tissue disease control group; 10 healthy persons were taken as normal control group at the same time. Using flow cytometer, recorded the mean fluorescence intensity of IL - 10R on the surface of T lymphocyte. Then perform statistic analysis for experimental results.ResultsThe mean fluorescence intensity of IL - 10R on the surface of T lymphocyte has no statistic difference between treatment and non -treatment SLE case groups (P =0. 096). The mean fluorescence intensity of IL - 10R on the surface of T lymphocyte in total SLE case group and RA case group has significant statistic difference compared with normal control group (P =0.006, P= 0.005 respectively). The mean fluorescence intensity of IL-10R on the surface of T lymphocyte in treatment and non - treatment SLE case groups were significantly different with normal control group (P =0.006, P =0.002 respectively) . The mean fluorescence intensity of IL - 10 R on the surface of T lymphocyte in total SLE case group was also obvious difference with RA case group (P =0.03). The mean fluorescence intensity of IL -10R on the surface of T lymphocyte in non - treatment SLE case group was also obvious difference with RA case group (P = 0.011). But themean fluorescence intensity of IL - 10R on the surface of T lymphocyte in treatment SLE case group has no statistic difference with RA case group (P=0.23). Undertake straight line correlation analysis between SLEDAI and the mean fluorescence intensity of IL - 10R on the surface of T lymphocyte in SLE. The correlation coefficient was -0.103, P =0. 51, which showed that there has no statistic difference between them.ConclusionSLE is a kind of disease which involved in many genes. Inheritance gene plays important role in the pathogenesis of SUE. Many researches have proved that IL - 10 increases in the body of SUE patients , and has related with the severity level of the disease and the genetic background. IL - 10R can also affect the function of cytokine. The results of our research demonstrated that the mean fluorescence intensity of IL - 10R on the surface of T lymphocyte in SLE case group is much higher than that of normal control group, which shows IL -10R in active phase SLE patients not only has gene polymorphism, but also higher than normal person at the protein level.
