Objective:Systemic lupus erythematosus (SLE) is a kind of autoimmunedisease caused by inherited genetic factors,gender,environmental factors and many other autologous-and-exoteric factors accompany with multi organs damage, characterized by the damaged function of suppressor T lymphocyte,the hyperactivation and proliferation of polyclonal B lymphocyte,generated antoantibody,immune complex develop immunologicmjury of tissues,it presents with various clinical manifestations.The pathogenesis of SLE is still unknown the etiological factors refer to heredity,environment abnormal of immunoloregulation,sexual hormone and other factors.The disease shows an obvious gender tendency, involving mainly the females, expecially those in childbearingyears.the rate of man and woman is about 1 to 9.To study the mechanism of estrogen receptor alpha,estrogen receptor beta synergistic effect in SLE,and relevance with Systemic Lupus Erythematosus Disease Activity Index and clinical,laboratory data,we designed this experiment and want to approach through the experiment:1. the expression difference of estrogen receptorα,estrogen receptorβin health adult and SLE patients 2. the expression difference of estrogen receptorα,estrogen receptorβin SLE patients male and female.3 the relevance among estrogen receptorα,estrogen receptorβand SLEDAI.4.the relevance among estrogen receptorα,estrogen receptorβand clinic data.5.the relevance between SLEDAI and parts of laboratory data.To offer some theory basis in elucidating pathogenetic,monitoring reactiveness,drug exploitation of SLE.Method:RT-PCR method is used to analysis the expression quantity of ERa,ERβin SLE patients of 40 cases and health adults of same number, The PCR products are separated by electrophoretic method through gelose with 4.0% ethidium bromide. The consequence is judged using gelose formatter and statistical analysis. Graded the 40 patients of SLE according to the SLEDAI-2000. analyze the expression difference of ERa,ERβand the correlation among SLEDAI,clinic data and laboratory data using the statistical methods.Result: 1.The expression level of ERa higher than normal control (P<0.05),but the expression level of ERβis lower than normal control in PBMC(P<0.05).2.The different expression level of ERa and ERβin PBMC of SLE patients in different gender has no statistics significance (P>0.05)3.The expression level of ERa of the patients with lupus nephritis higher than the patients without lupus nephritis(P<0.05).4.The ERP expression level of patient with angiocardiopathy is lower than the patients without angiocardiopathy(P<0.05).5.The expression of ERa and ERP in SLE patients have no statistical significance(P > 0.05),but the ERβexpression and the SLEDAI has a negative correlation (P<0.05).6.The SLEDAI has a reciprocal ratio with RBC (P<0.05).Conclusion:1.The expression of ERa and ERP in PBMC of SLE patients is elevated than normal control which suggested that ERα,ERβmay take part in the pathology of SLE.2.The ERP expression in PBMC has a negtive correlation with SLEDAI which suggested that ERP may be related with the activity of SLE.3.The SLE patients with Lupus nephritis has a higher ERa expression than the patients without Lupus nephritis,shows a important role in Lupus nephritis.4.The SLE patients with angiocardiopathy has a lower ERP expression than the patients without angiocardiopathy,shows the ERβhas a protection in cardiovasculair.5.The SLEDAI has a reciprocal ratio RBC,the alteration of RBC can be a indicator of SLE activity.
