| Osteoporosis is a kind of disease with the characteristic of reducing with bone amount, changing the micro-structure of bone and increasing the danger in fracture. Its incidence of disease is high, dangerous , have already stood side by side with artery sclerosis, high blood pressure, diabetes, tumour,etc. for old people's important disease at present. Western medicine come with estrogen, calcium pharmaceutical, calcitonin for treating . Although there is certain curative effect, there are greater side effects. So there are greater clinical meanings in preventing and curing the disease with traditional Chinese medicine .Professor Jiangliang Duo .according to clinical experience ,spend the Sanxian soup (barrenwort, celestial cogongrass, hairyvein agrimony)for many years in order to treat climacteric syndrome and improve woman symptom of climacteric . The soup effects emarkably. The earlier stage of the experiment indicates that the Sanxian soup promoted the proliferation of osteoblast-like cells. So we imagine through the experiment of setting up animal model and training external osteoblast-like cells to combine together in order to offer the experiment basis for clinical practice ,probe the function mechanism of traditional Chinese medicine to prevent and cure Osteoporosis. Concrete experiment content is:First methods:1. Observe the Sanxian soup to influence osteoblast-like cells UMR106 and study relevant enzyme ( ALPase) in the osteoblast-like cells' decomposition and clear liquid.2. Select 59 Wistar rats aged three months and make the operation of variotomy .Then divide into false operation group, model group, positive group, Sanxian low dose group. Sanxian middle dose group and Sanxian high dose group. The false operation group and model group were given physiological saline solution once a day. Thepositive group was fed Nilestrioli once a week. The Sanxian low dose group was fed Sanxian soup 5.98g-kg~L d"1, the Sanxian middle dose group 11.96 g-kg^d"1 and the Sanxian high dose group 23. 92 g-kg~Ld~' for twelve weeks. At the end of the twelfth week ,all rats were sacrificed .We determine rats' calcium, phosphorus, ALPase in the serum ,urine calcium, urine phosphorus and bone mineral density.Two results :1. The Sanxian soup of the appropriate concentrations(Img/L, 5mg/L, lOmg/L, lOOmg/L, 250mg/L, 500mg/L, lOOOmg/L) promoted the proliferation of osteoblast-like cells .The concentration (lOmg/L) promoted the proliferation of osteoblast-like cells better than 100 mg/L, 250 mg/L, 500 mg/L, 1000 mg/L and have remarkable difference compared with the groups. The concentration between l-10mg/L had an increasing tend on promoting the proliferation of osteoblast-like cells. In the osteoblast-like cells' decomposition liquid , the concentrat ion( 1 mg/L, lOmg/L) a little increased the ALPase activity in the cells more compared with the blank group. In the osteoblast-like cells' clear liquid, the concentration (Img/L) a little increased the ALPase activity in the cells compared with the blank group.2. The situation of model group : Calcium and phosphorus in the serum obviously reduced than the false operation group( P < 0. 01 or P < 0. 05). Urine calcium obviously raised than the false operation group ( P < 0. 05). Urine phosphorus increased than the false operation group to some extent ( P> 0. 05). The ALPase in the serum rised than the false operation ( P> 0. 05). Bone mineral density notably reducd than the false operation group (P < 0.05). All these shows that the model was made sucessfully.
