Objective: The aim of the study was to observe the effects of Bushenfumai(qianjin) liquor on the releasing of Nitric Oxide(NO),the activity of Nitric Oxide Synthase (NOS) and the expression of iNOSmRNA in heart and brain of the hypoxic rat.Method: The hypoxic rats were randomly divided into four groups, the blank (normoxia) group, the model(hypoxia)group, western medicine group (hypoxia+ Vitc) and the Chinese medicine group (hypoxia+ Bushenfumai liquor). Expect the blank group, the other groups were exposed to hypoxia(10%O2) for six hours everyday, lasted two weeks. We got the homogenates of organization from the killed rat's heart and brain, and determined the releasing of NO with nitric acide reducing-enozyme method, and determined the level of activity of NOS by absorbance under the 530nm wave length. Then the total RNA was extracted from the homogenates with the method of guanidineium thiocynate. Finally, the reverse transcription and polymerase chain reaction(RT-PCR)were performed on every group. RT-PCR products were fractionated in to size through a 1.0% agarose gel, electrophoresis bands with ethidium bromide were visualized under the ultravio!et(UV) light and photographed. Bands on film were scaned by the densitometer, and then were quantified on computer by the image analysis software. Then the iNOSmRNA level were determined.109Results:(l)Both the NO level of the heart and brain of the hypoxia group increased significantly than the blank group , P<0.001(P=0.000), P< 0.05(P=0.038) . Both the NO level of the heart and brain of the Chinese medicine group decreased significantly than the hypoxia group P < 0.001(P=0.000) and the Chinese medicine group decreased more significantly than the western medicine group, P<0.01(P=0.008) s P< 0.001(P=0.000).(2) There was no significant difference between the hypoxia group and blank group of the NOS activities of the heart and brain P>0.05(P=0.210) > P>0.05(P=0.817) . The Chinese medicine group increased the heart NOS activities than the hypoxia group P<0.05 (P=0.026). There was no significant difference among other groups.(3) Both the expression of the iNOSmRNA of heart and brain of the hypoxia group increased significantly than the blank group P < 0.001(P=0.000) , P < 0.001(P=0.000). Both the expression of the iNOSmRNA of the heart of Chinese medicine group and western medicine group decreased significantly than the hypoxia group, P<0.001(P=0.000) , P<0.001(P=0.000), and Chinese group was lower than western group, PO.05 (P=0.022) . Both the expression of the iNOSmRNA of the brain of Chinese medicine group and western medicine group decreased significantly than the hypoxia group, P<0.001(P=0.000) ^ P<0.05(P=0.039), andChinese group was lower than western group, P<0.05 (P=0.014) .Conclusion: The Chinese medicine Bushen Fumai (qian jin) liguor decreased the NO and the iNOSmRNA level of hypoxia rat's heart and brain, as showed that Bushen Fumai (qian jin) liquor had a significantly protective effect on the hypoxia rat s' heart and brain.
