Syringa pubescens is a shrub with a wide distribution in China. Its flowers have long been used to treat chronic hepatitis, enteritis, cancer of the esophagus, cirrhosis,and cancer of the liver among the people. But the medical use of its flowers has notbeen officially developed to date. In our previous work, we examined the water-solubleconstituents of this plant and determined the n-BuOH position as the major anti-cirrhosis position. Studies on the chemical constituents of the n-BuOH position resultedin the isolation of two secoiridoid glucosides: oleuropein and oleoside 11-methyl ester.In this paper, we made a thorough pharmaceutical-chemistry investigation and study onthe anti-cirrhosis n-BuOH position. Our short-term achievements are established asfollows:1. Through perpendicular experiments, the optimum technological conditions weredetermined as this: adopting reflux mode, the initial extraction solvent using 90%industrial ethanol, the average reflux time being 3 hours and the first extraction so1ventbeing cyclohexane-ethyl acetate (2:8). Later, the n-BuOH ointment was disposed intopowder after transforming the rearmost technology. As a result, the extraction ratio of the raw medicine (namely "XiaoYeDingXiangSu" ) is fairly good (about 9.5%). ByHPLC detection, the content of the main effective constituent-oleuropein in thesamples has all exceeded 88%.2. According to Chinese Pharmacopoeia and the Study Index of New TraditionalChinese Medicine, We not only studied the content limitation, identification, acidity,solubility,drying test, quantitative analysis, and stability testing of the semi-finishedmedicine, but also established the quality standard of the raw medicine along withdrafting explanation. To quantitative analysis, the crux part of qua1ity control, we madeuse of RP-HPLC which has high accuracy and studied the methodology of it.3. The isolation and structure elucidation of a flavonoid-type glucoside namedkaempferitrin is reported:These works fill in the gaPs Of the ndsuOH position in the phannaceuical-chemistry field, and are bound tO exert an imPortat influence on the development oftw pobW the formulation of a new anti-cirrhosis drug and the moddriZationof the traditional Chinese medicine.
