In order to study the anti-cirrhosis effective constituents and consider modern separation techniques for effective intermediate components in depth, including extraction process and quality standards, the fingerprint of Syringa pubescens Turca needs to be setted up, so it provides the scientific evaluation for anti-fibrosis on the production of health products.Under the guidance of Pharmacology, To establish the HPLC-NNE13C-NMR of Syringa pubescens Turca extract, six kinds of Macroporous Absorption Resin are selected to optimize extraction process by the orthogonal method, and the content of oleuropein from a new medicinal herb Syringa pubesceus Turca was determined by RP-HPLC. Consequently, the best extraction process is established. There are good linear relationship in this method and it can be used scientifically to evaluate the quality of different parts of Syringa pubescens Turca both qualitatively and quantitatively. In this subject the chromatographic conditions included: KromasilC18 (4.6mm×250mm,5μm), the flow rate was 1.0mL﹒min -1. The column temperature was 25℃.The gradient elution was adopted with acetonitrile and water(25∶75) as a mobile phase. The detection wavelength was at 285 nm. For further to compare other active ingredients with the standard reference oleuropein quantitative the carbon peak height ratio of HPLC-NNE13C-NMR to establish the quantitative calculation of the relationship. The results show that, This method is feasible as an effective quality control of new technologies. Compared to other fingerprinting techniques in current for the evaluation of product quality and stability, a reasonable assessment of the production process are more reliable and comprehensive. On the basis of the best extraction process, in this issue the column chromatography silicagel, thin-layer chromatography and HPLC are used repeatedly, the purification technology of the anti-cirrhosis effective constituents from Syringa pubescens Turca is separated detailly. Four compounds are gained, they are oleuropein(1),(8Z)-ligstroside(2), echinacoside(3) and D-mannito(4), the compounds 2 and 3 are firstly separated form Syringa pubescens Turca.
