Hanning Model Of Rats With Blood Stasis Syndrome Peptic Ulcer And Gastric Mucosal Blood Flow, Blood Rheology Observed

Purpose:To establish the rats model of peptic ulcer with the syndrome of blood stasis due to cold accumulation, which is for clinical research.Material and method:20 rats were randomly divided into two groups :the control group(n = 10) and the model group(n = 10) . The model group was induced by improved cold exposure, which was by intragastric administration with indometacin at the same time. No cold exposure was to the control group. The primary outcome measures were the syndromes, sighs, gastric mucosa, and gastric mucosal blood flow and hemorheology.Results:Model rats appeared hunched,lazy, unresponsive, chill,cluster, breathing faint, hair without luster, less water quantity , clear urine, wet and lousy stools, dark purple tails and claws, dark red ears. The rats of Control group were vital, strong, agile, feeding well, hair with luster, robust. Through the microscope, gastric mucous of the rats in control group was almost normal. On the contrary, the gastric mucosal damage of model rats was evident, for example striped erosion which was mostly in the body of stomach, cell denaturation in the whole mucous membrane, and lost of gastric pits. Besides, both swelling, necrosis or amotio of the chief cell and mucous cell, focal hemorrhage and acute inflammatory reaction in membrana propria, and damage below the gastric mucus were the characteristics of the gastric mucosal damage of model rats. As compared with control group, in model group, ulcer index(11.55±2.74)and hemorheology were increased remarkably. Meanwhile, gastric mucosal blood flow in model group(57.18±7.12)was decreased obviously (P<0.01) .Conclusion:The rats model of peptic ulcer with the syndrome of blood stasis due to cold accumulation was established by improved cold exposure and indometacin method. This new animal model was suitable for syndrome of traditional chinese medicine and the characteristics of peptic ulcer with the combination of disease and syndrome,which was for the experimental research of traditional chinese medicine.
