Integrative thinking is the basic thinking model of Traditional Chinese Medicine (TCM) in diagnosing and treating disease. But current efficacy evaluation methodologies of TCM mostly copy the methods of western medicine, using single or a handful of pathological indexes' changes to evaluate the efficacy of TCM. This is logical for western medicine, whose primary mechanism is single pathway and single target point. But it violates integrative thinking theory of TCM, difficult to objectively reflect actual treatment efficacy of TCM. The feature of TCM treating disease is multiple pathways and multiple target points, emphasizing integrative coordination of organism. Using the efficacy evaluation methodologies of western medicine to evaluate TCM often leads to the efficacy evaluation of TCM lacking comprehensiveness and objectivity, and not accord with basic theoretical principle of TCM treating disease, because TCM usually works slowly and its treatment target points are always not clear. Therefore, it is maybe more suitable for the thinking theory of TCM to explore an evaluation methodology based on the whole functional changes of organism, which is a supplement and optimization for current TCM efficacy evaluation modals.Metabonomics, as an important branch of system biology, is aimed at researching the metabolic pathways in living systems by means of investigating the integrated changes of metabolite in organisms. This is unified with the integrative treatment thought of TCM, which provides a great technology platform for evaluating TCM efficacy from the global aspect.In this article, blood stasis animal models were established by means of ice-water stimulation and adrenaline hydrochloride subcutaneous injection, the efficacy of Compound salvia tablets to blood stasis animals was evaluated in the aspects of pathological experiments and metabonomics. The main work was as follows:1. Blood stasis rats model were established, whose hemorheology indexes were detected to evaluate the efficacy of Compound salvia tablets to blood stasis rats. The results showed that, compared with the control group, the model group rats' hemorheology indexes, those were blood viscosity, plasma viscosity, hematocrit and fibrinogen content, were all obviously increased, the differences were significant (p<0.05) or extremely significant (p<0.01). This indicated that it was able to replicate blood stasis model successfully by means of ice-water stimulation and adrenaline hydrochloride subcutaneous injection. Compared with the model group, plasma viscosity and fibrinogen content of the Aspirin group, plasma viscosity, hematocrit and fibrinogen content of the low dose group of Compound salvia tablets, blood viscosity (when shear rate was 1.00s-1 and 200.00s-1 respectively), plasma viscosity and fibrinogen content of the high dose group of Compound salvia tablets improved obviously, the differences were significant (p<0.05) or extremely significant (p<0.01). The differences of the four indexes between the low dose group of Compound salvia tablets and the Aspirin group were not significant. The improving effect of high dose of Compound salvia tablets on blood viscosity, plasma viscosity and fibrinogen content was better than Aspirin enteric-coated tablets, the differences were significant (p<0.05) or extremely significant (p<0.01). The experiment indicated that Compound salvia tablets were able to effectively inhibit the influence of blood stasis on rats' hemorheology indexes. The efficacy of low dose of Compound salvia tablets was similar to Aspirin enteric-coated tablets, but the high dose had better efficacy.2. Blood stasis mice model were established, whose coagulation time was detected to evaluate the efficacy of Compound salvia tablets to blood stasis mice. The results showed that, compared with the control group, the model group mice's coagulation time was obviously shortened, the difference was extremely significant (p<0.01). Both of Aspirin enteric-coated tablets and Compound salvia tablets were able to effectively inhibit the influence of blood stasis on mice coagulation time, compared with the model group, the differences were extremely significant (p<0.01). But the difference between Compound salvia tablets and Aspirin enteric-coated tablets was not significant. The experiment indicated that Compound salvia tablets were able to effectively prolong coagulation time of blood stasis mice, and its efficacy was similar to Aspirin enteric-coated tablets.3. A plasma metabonomics method based on the gas chromatography combined with mass spectrometry (GC-MS) was used to evaluate the efficacy of Compound salvia tablets to blood stasis mice. The results showed that, compared with the control group, the model group mice's plasma metabolic profiling changed obviously. L-alanine, L-valine, malic acid and arachidonic acid in the model group were significantly increased, the differences were significant (p<0.05) or extremely significant (p<0.01). Both of Aspirin enteric-coated tablets and Compound salvia tablets were able to effectively inhibit the influence of blood stasis on L-alanine, L-valine, malic acid and arachidonic acid in mice plasma, compared with the model group, the differences were significant (p<0.05) or extremely significant (p<0.01). Compared with the Aspirin group, only arachidonic acid in the Compound salvia tablets groups significantly decreased (p<0.01). The differences of L-alanine, L-valine and malic acid between the Compound salvia tablets groups and the Aspirin group were all not significant. The experiment indicated that, Compound salvia tablets were able to effectively improve the changes of metabolite in blood stasis mice plasma; for arachidonic acid, the efficacy of Compound salvia tablets was better than Aspirin enteric-coated tablets; but for L-alanine, L-valine and malic acid, Compound salvia tablets and Aspirin enteric-coated tablets had similar efficacy.These results indicated that, the research result of metabonomics was consistent to the synthesis results of pathological experiments, and it was able to reflect living systems' metabolic state more comprehensively. Metabonomics had great application value in the study of TCM integrative efficacy evaluation.
