Objective:Researching to compare difference and similarity between Volatile oil component and flavonoid of Patchouli in Vietnam and China, finding the change rule of Patchouli component after many years of planting. In order to provide reference document to Patchouli planting in Vietnam and China; concurrently through researching comparison of Volatile oil content and total flavonoid after using separated technology and processing many kind of patchouli having different origin, selecting the most appropriate separated technology and processing to ensure the quality of Patchouli decoction pieces.Method:Gather method to extract volatile oil, using GC-MS (Gas Chromatography- Mass Spectometry) method to search and detect components, compare similarity and difference in their content and component;Using ultraviolet spectrophometric method to establish the method assay total flavonoid components with Apigenin and Rutin used as the reference substances. Concurrently using HPLC (High Performance Liquid Chromatography) method to search and compare of similarity and difference of flavonoid group and finger printing of the methanol extraction part. Investigate the relative of non-volatility in Patchouli from two contries.Examine factor which can impact to change, making a processing and separated technology basing on the key target-Volatile oil content and flavonoid, finding the most appropriate separated technology and processing.Result:Through analysis and comparison of volatile oil content and characteristic fingerprint of patchouli in two countries, the result of fingerprint similarity is quite low, distinction includes Guangzhou- Guangdong-China 0.8018; Zhanjiang- Guangdong 0.8458, Gaoyao-Guangdong 0.9088, Zhixin company-Guangdong 0.8687; Ha noi1 0.8345, Ha noi2 0.9207, Ha noi3 0.8666, Viet Tri 0.9126, Son La 0.9154, Vinh 0.8800; Zhanjiang GAP depot1 0.7746, Zhanjiang GAP depot2 0.7650。Difference of content multi- compose is quite big, content of Vietnam patchouli volatile oil is correlative in turn as Hanoi1 1.52%, Viet Tri 1.38%, Vinh 1.38%, Son La 1.89%, Ha noi2 2.07%, Ha noi3 2.16%, Viet Tri 1.38%, in general, volatile oil content in these areas is higher than in Guangdong (China)-1.32%, Zhanjiang Guangdong 1.43%, Gaoyao- Guangdong 1.71%, Zhixin company- Guangdong 1.02%.The total content result of total flavonoid is correlative in turn as China 0.82%, Vietnam 0.95%(Apigenin); China 0.71%, Vietnam 0.78%(Rutin).Establishing HPLC fingerprint of total flavonoid substance group of many kinds of patchouli having different origin in Vietnam and China, first step,13 general peaks are set up as 13 specific peaks, the similarity includes:Guangzhou- Guangdong- China 0.9450; Zhanjiang- Guangdong 0.9448, Gaoyao- Guangdong 0.9088, Zhixin company- Guangdong 0.8924; Ha noi 1 0.9449, Ha noi20.9377, Ha noi30.8772, Viet Tri 0.8782, Son La 0.8770, Vinh 0.9513; Zhanjiang GAP depot1 0.6411, Zhanjiang GAP depot2 0.5593.The substance groups of types of patchouli having different origin, which are extracted by methanol, have fingerprint similarity in turn as:Guangzhou-Guangdong-China 0.9631; Zhanjiang- Guangdong 0.9451, Gaoyao- Guangdong 0.9342, Zhixin company- Guangdong 0.8912; Ha noi1 0.8439, Ha noi2 0.9377, Ha noi3 0.8772, Viet Tri 0.8782, Son La 0.8673, Vinh 0.9213; Zhanjiang GAP depot1 0.762, Zhanjiang GAP depot2 0.7593.Processing method no.1 and no.2 have the highest volatile oil content with 1.20±0.01%, the difference has statistical meaning, therefore, the most appropriate processing method and separated technology which raise volatile oil content up are method no.1 and technology no.2, this means drying daily and keeping until becoming dry nightly. After that each lkg of medicine will be sprinkled with 200ml of water, will be steeped in, cut into sections, then we get patchouli decoction pieces immediately.Conclution:Volatile oil content of patchouli in Vietnam is higher than in China; volatile oil component combination of patchouli in two countries has the fixed similarity, however, there is a big difference in content, and this proves that if patchouli leave have different origins, their combinations are different.Applying Apigenin as reference substance, through using the refined Poliamide and then using direct ultraviolet spectrophotometry method to quantify total flavonoid in patchouli, this method is simple, fast and has a good linear relationship, high sensitivity, accuracy, suits to definition total flavonoid in patchouli. The dosimetry result shows that total flavonoid content of patchouli in Vietnam is clearly higher than introduction patchouli in China.Establishing fingerprint HPLC of total flavonoid., implementation of this method is simple, repeated; this method can be used as target to assess and evaluate quality of total flavonoid of Patchouli. Patchouli from different origins have its specific finger print of total flavonid has fixed differences After patchouli was introduced to Guangdong China, because of ecological environmental condition, the content of mainly effective components changed clearly.When patchouli is processed, it needs to control closely operation of fermenting technology, separated technology which impact negligibly to content of volatile oil and total flavonoid.
