| Radix Phytolaccae has diuresis, decreasing swelling and detoxication, precluding knot traditional effects. It often cures Renal edema, Cardiogenic edema and Ascites of cirrhosis of liver in clinic. But now its effective part or components of diuresis has not been found. In this paper the methods of the natural pharmaceutical chemistry and pharmacology were used to search for diuretic effective part or components of Radix Phytolaccae. The main researches were aimed to identify the variety of the traditional Chinese medicine, to produce the different solvent extract, to carry out the pharmacological experiment of diuresis, to set up HPLC spectra of Phytolacca amencana L. and the parts which was associated with diuresis, to extract and isolate the alkaloids of Phytolacca americana L., to analysize the liposoluble components by GC-MS. Result: The mixture of raw alkaloid and raw saponin was the diuretic effective part. The HPLC spectra of Phytolacca americana L. and the parts which is associated with diuresis were set up. Two compounds of alkaloid were isolated. Eight liposoluble components were identified.To identify the variety of the traditional Chinese medicine: Phytolacca americana L. is the variety in this topic through the characters identification, transverse section and medicine powder microidentification, specific qualification tes and TLC.To determine the diuretic effective part: Firstly, three dosage groups (10, 20, 40 time) were set up. According to effect of each group on healthy mouse's urine, 20 time dosage was the effective dosage by the aid of statistic analysis, which including control group (0.9%salt water), positive comparison group (hydrochlrothiazide, intermediate effective medicine used in clinic). Secondly: The extract of Phytolacca americana L. were separated into severalparts according to the different polarity of solvents: 1-CO2-2, 1- n-butanol-3, 1-water-4, 1-ash-5. According to effect of each part on healthy mouse's urine, 1-n-butanol-3 was the effective part and 1-ash-5 has some diuretic action by the aid of statistic analysis, which including control group, positive comparison group. Thirdly: The rest of 1-n-butanol-3 was divided into three parts (2- n-butanol-l,2-n-butanol-2, 2-n-butanol-3). The part of 2-n-butanol-2 was separated into three parts according to the different polarity of solvents: 3-ether-l, 3-acetic ether-2, 3-n-butanol-3. According to effect of each part on healthy mouse's urine, the part of 3-n-butanol-3 had some diuretic action, by the aid of statistic analysis, which including control group, positive comparison group, but its action had no statistical difference. Finally: The part of 2-n-butanol-3 was separated into two parts according to the chemical characters of the main components: raw alkaloid (4-alkaloid-l),raw saponin (4-saponin-2). And the mixture of 4-alkaloid-l and 4-saponin-2 was tagged 4-minture-3. According to effect of each part on healthy mouse's urine, the part of 4-minture-3 was the effective part by the aid of statistic analysis, which including control group, positive comparison group.To set up HPLC spectra of Phytolacca americana L. and the parts which were associated with diuresis: HPLC spectra of Phytolacca americana L. and the parts which were associated with diuresis were set up by using RP-HPLC and solvent gradient elution (acetonitrile, water).
