Objective To investigate the therapeutic effect of selenium enriched traditional Chinese medicine on chronic nonbacterial prostatitis in rats.Methods Fifty healthy adult male SD rats were selected,weighing about 250 g,and randomly divided into 5 groups.The method for preparing a rat model of chronic nonbacterial prostatitis by Wei Wuran was followed,and according to the preparation standard of chronic nonbacterial prostatitis animal model.Except the rats in the blank control group,the remaining 4 groups were anesthetized with 7% chloral hydrate(0.5ml/100g)by intraperitoneal injection,then castration surgery was performed under sterile conditions,the skin was sutured,disinfected and bandaged,returned to the squirrel cage,and free diet.Starting from the second day after operation,SD rats were injected subcutaneously with 0.25 mg / kg·d of estradiol benzoate into the back for 30 days.After the model was successfully prepared,the four groups of SD rats were divided into four groups according to the treatment methods: normal saline treatment group(model group),selenium-free traditional Chinese medicine treatment group(traditional Chinese medicine group),Low dose selenium enriched traditional Chinese medicine treatment group(low selenium group),high dose selenium enriched traditional Chinese medicine treatment group(high selenium group),5animals in each group of blank control group and model group,and 10 animals in each other groups.The four groups were treated by intragastric administration(1ml/100g)for 14 days.The contents of inflammatory factors TNF-?,IL-1 and i NOS in prostate tissues were measured by ELISA,and pathological sections of prostate tissue were prepared.Results After 14 days of gavage treatment,the contents of TNF-?,IL-1 and i NOS in prostate tissue of SD rats in model group were significantly higher than those in other groups,the difference was statistically significant(P<0.05);at the same time,under the pathological section microscope,the gland lumen was convex and narrow,without obvious secretion,with obvious inflammatory cell infiltration.The contents of TNF-?,IL-1,i NOS in prostate tissueof the Chinese medicine group was lower than that of the model group,and the expression of TNF-?,IL-1,i NOS was decreased,the difference was statistically significant(P < 0.05),and the pathological section of the group showed that the inflammation was reduced.Compared with the traditional Chinese medicine group,the contents of TNF-?,IL-1,i NOS in the prostate tissue of the rats in the selenium enriched traditional Chinese medicine group decreased,the expression decreased,the difference was statistically significant(P < 0.05),among them,the high selenium group decreased more significantly than the low selenium group,and the pathological section showed that the inflammation improved better.Conclusion Compared with the treatment effect of selenium-free traditional Chinese medicine,selenium enriched traditional Chinese medicine can improve the inflammatory response of chronic nonbacterial prostatitis,reduce the contents of TNF-?,IL-1,i NOS,and the therapeutic effect of high selenium traditional Chinese medicine is better than that of low selenium traditional Chinese medicine and selenium-free traditional Chinese medicine in the treatment of chronic nonbacterial prostatitis.
