Squamous cell carcinoma (SCC) is common malignant tumor of the oral and maxillofacial region, and the SCC of tongue had been the first place among the malignant tumor. Tongue cancer usually has serious malignancy and invasion, high growth velocity, and in the advanced stage the tongue will be fixed by the cancer spreading to mouth floor and mandible. High metastatic frequency and redundant lymph vessel and blood circulation lead to the quality of life of patient seriously. Now operation is the main choice. But the patient often has a bad quality of life after the operation because of high post-operation recurrence rate.Nowadays, it was hot spots to use the effective component that were distilled from plants to suppress tumor cell's multiplication, induce differentiation and prompt cell apoptosis. It had been reported that IAA and HRP had the function of anti-tumor. But it hadn't been studied whether IAA and HRP could inhibit the growth of SCC of tongue. So this research was to explore the effects of IAA and HRP inducing the differentiation of Tca-8113 cell line and its mechanism.Objective: Investigate the effects and mechanisms of IAA/HRP on Tca-8113 cell line.Materials and Methods:1. Tca8113 cell line was provided by Shanghai Second Medical University.2. IAA and HRP was bought from Genesis Biotechnology Inc.3. Growth condition and morphology of the cells after treated by IAA/HRP were observed under inverted microscope.4. Tca-8113 cells were treated with IAA in concentration of 0μmol/L, 20μmol/L, 40μmol/L, 60μmol/L, 80μmol/L,100μmol/L, respectively, at the same time with HRP in concentration of 1.2mg/L. And the OD of the cell line was detected after 24h, 48h, 72h, 96h by MTT method, then we calculated inhibition ratios of each group.5. Distribution of cell cycle was detected by flow cytometer (FCM).6. Corpuscular microstructures of experiment group and control group were observed under transmission electron microscope (TEM).7. Immunochemical method was used to detect the change of the expression of Bcl-2.Results:1. Treated Tca-8113 cells with IAA/HRP in different concentrations for the same time, the deviation of depressant effect between experiment group and all the control group had statistical significance (P<0.05), the inhibitory rate in the same concentration of IAA/HRP had statistical significance in different time, compared with all the control group (P<0.05).2. The proportion of G2/M and S phase went down distinctively while that of G0/G1 with a rising trend (P<0.05).3. Immunochemical results indicated that IAA/HRP decrease the expression level of the Bcl-2.Conclusions:1. IAA/HRP can inhibit the growth of Tca-8113 which is concentration- and time-dependent.2. It has significant deviation in distribution of cell cycle between experiment group and control group, the proportion of G2/M and S phase went down distinctively while that of G0/G1 with a rising trend. Therefore, IAA/HRP inhibits the proliferation of Tca-8113 cells in vitro, the mechanism of which may be involved in cell cycle arrest of the cells.3. IAA/HRP can decrease the expression level of the Bcl-2, it maybe enhance apoptosis.4. The results showed that: IAA had Inhibition Effect on Tca-8113 cell line for over 60μmol/l.
