Esophageal squamous cells carcinoma is one of the commonest malignant tumors in our country.Esophageal carcinoma morbidity and mortality rank first in China.Esophageal cancer chemotherapy is the main means of treatment,Multi-drug resistance is the main reason for the failure of chemotherapy.At present almost known anticancer drug resistance can occur,the tumor drug resistance has become the focus of the study.Glutathione S-transferase(GST-π),GST-π-also known as the placenta or acidic isoenzyme,located in human chromosome 11 q13,gene structure for the length of 3 kb,its molecular weight of 55 KD.GST-πwidely distributed,rich content, commonly found in red blood cells,lung,breast,prostate,placenta,and other normal tissues.GST-πis expression in a variety of human cancer tissues.GSH tumor cells by regulating the level of increase in GST activity,thereby speeding up drug metabolism and chemical resistant.One,GST-πis the main cause of resistance,was considered a sign of cancer.Topoisomerase(TOPO-Ⅱ),also known as the gyrase,TOPO-Ⅱthere are two types of isozymeⅡa andⅡβ,molecular weight were 170 KD and 180 KD, respectively located in human chromosome 17 q21-q22 and human chromosome 3 p24.TOPO-Ⅱtwo isozyme widely expressed in tumor tissue,no tissue-specific.Their resistance mainly through the mechanism of DNA-mediated response to the fracture and a DNA-enzyme complex,various chemotherapy drugs through more complex and affect the stability of the proliferation of cancer cells.Study found that malignant tumors in a variety of people in multi-drug resistance gene GST-πand TOPO-Ⅱ,and so there are a total of expression.In esophageal cancer in the GST-πTOPO-Ⅱand the expression at home and abroad have not reported.To explore the GST-πand TOPO-Ⅱin squamous cell carcinoma of the esophagus and its significance,this study using immunohistochemistry SP detected GST-πand TOPO-Ⅱin esophageal squamous cell carcinoma of the protein expression of GST-πand TOPO-Ⅱexpression and the expression and the relevance of esophageal cancer.Materials and methods:1 Sample:48 cases of esophageal squamous cell carcinoma were collected in the HeHan Tumour Hospital and the First Affiliated Hospital,Zhengzhou University from September 2004 to November 2006.All the cases had no histroy of the chemotherapy, radiotherapy and immunotherapy.2 Using the technology of immunohistochemistry SP to detect the expression of GST-πand TOPO-Ⅱin the 48 cases of esophageal squamous cell carcinoma,30 cases of normal mucosa of esophageal respectively.3 Statistical analysis:All the dates were analyzed by SPSS 11.0 statistical package,the count information calculated the positive rate,The comparison of positive rates uses the Chi-square;The relation of two variable are analyzed by the spearman level correlation analysis.The level of significant difference is a=0.05.Results:1.Immunohistochemical results indicated:the positive expression rate of GST-πprotein in normal mucosa of esophagus is 30%(9/30);The positive expression rate of GST-πprotein in the carcinoma tissues is 64.58%(31/48),the positive expression rate of GST-πprotein in the carcinoma tissues is compared with that normal mucosa tissues from each other,There is stastital significance(P<0.05).2.Immunohistochemical results indicated:the positive expression rate of TOPO-Ⅱprotein in normal mucosa of esophagus is 20%(6/30);The positive expression rate of TOPO-Ⅱprotein in the carcinoma tissues is 41.67%(20/48),the positive expression rate of TOPO-Ⅱprotein in the carcinoma tissues is compared with that normal mucosa tissues from each other,There is stastital significance(P<0.05).3.In the 14 cases of metastatic group,the positive rates of expression of GST-πprotein in the tumor tissues are 64.28%(9/14);in the non-metastatic group, they are 64.71%(22/34).There is no significant difference between the two groups (P>0.05).4.In the 14 cases of metastatic group,the positive rates of expression of TOPO-Ⅱprotein in the tumor tissues are 42.86%(6/14);in the non-metastatic group, they are 41.18%(14/34).There is no significant difference between the two groups (P>0.05).5.In the tissues of esophageal squamous cell carcinoma,the positive radio of expression of GST-πprotein on the well-differentiated carcinoma,moderately differentiated carcinoma and poor differentiated carcinoma is 73.33%(22/30) 57.14%(8/14) and 25.00%(1/4) respectively.There is a significant difference between the two groups(P<0.05).6.In the tissues of esophageal squamous cell carcinoma,the positive radio of expression of TOPO-Ⅱprotein on the well-differentiated carcinoma,moderately differentiated carcinoma and poor differentiated carcinoma is 33.33%(10/30),50%(7/14) and 75%(3/4)respectively.There is a significant difference between the two groups(P<0.05).7.The relativity of the expression of GST-πand TOPO-Ⅱwas analysized by statistics,Results showed that the both are negative correlation(r=-0.787,P<0.05).Conclusions:1.The positive rates of the expression of GST-πand TOPO-Ⅱprotein in squamous carcinoma tissues of esophagus and normal mucosa tissues were low in turn,Therefore the comparison of the expression of GST-πprotein among carcinoma tissues and normal mucosa tissues has statistical signicance.It indicaed that GST-πand TOPO-Ⅱparticipated in occurrence and development of esophageal cell carcinoma.2.The expression of GST-πand TOPO-Ⅱin esophageal squamous cell carcinoma tissues with lymph node metastasis and in without lymph node metastasis were no statistical signicance.It indicates that there were no correlation between the expression of the GST-π,TOPO-Ⅱproteins and metastasis of esophageal cell carcinoma.3.There was different expression level of GST-πand TOPO-Ⅱin the different differentiation of esophageal squamous cell.With the increased level of tumor differentiation,the expression of GST-πwas increased.TOPO-Ⅱwas reversed.It indicates that there were correlation between the expression of the GST-π,TOPO-Ⅱproteins and differentiation of esophageal cell carcinoma.4.The expression of GST-πand TOPO-Ⅱprotein in esophageal squamous cell carcinoma was negative correlation by statistical analysis,which indicated that there may be different resistance mechanisms between GST-πand TOPO-Ⅱ.
