ObjectiveTo observe the effect of catalpol,berberine and their combination on the expression and the secretion of TNF-α,IL-1βand IL-6 in insulin resistant 3T3-L1 adipocytes so as to reveal the scientific connotation of Rhizoma Coptidis Pill and the effect mechanism of catalpol,berberine and their combination in improving insulin resistance by inflammatory factor channels.MethodsPart one Effect of Berberine Catalpol and Their Combination on IR 3T3-L1 Adipocytes glucose consumputionAdipocyte IR model was induced with dexamethasone, and divided into model group and treatment groups which were treated with catalpol, berberine, and combination of catalpol and berberine respectively. Normal control group was also designed. Glucose consumption of adipocytes incubated with corresponding medicine was assayed through the glucose oxidase method. Part two Effects of Berberine Catalpol and Their Combination on the Secretion and mRNA Expression of TNF-α,IL-1β,IL-6 in IR 3T3-L1 AdipocytesCollect the cells and supernatant of culture solution, then the secretion of NF-α,IL-1βand IL-6 was detected by enzyme linked immunosorbent assay, and the mRNA expression of TNF-α,IL-1βand IL-6 was measured by using Real time PCR.ResultsPart one Effect of Berberine Catalpol and Their Combination on IR 3T3-L1 Adipocytes glucose consumputionCompared with control group, the glucose consumption of model group was lowered significantly (P<0.01),whereas the glucose consumption of catalpol group, berberine group and combination of catalpol and berberine group were significantly improved compared with model group (P<0.01); In addition, the effect of combination group was better than that of catalpol group as well as berberine group (P<0.01).There was no significant difference between the effect of catalpol group and berberine group(P>0.05).Part two Effects of Berberine Catalpol and Their Combination on the Secretion and mRNA Expression of TNF-α,IL-1β,IL-6 in IR 3T3-L1 AdipocytesThe expression and secretion of TNF-α,IL-1βIL-6 in model group increased significantly compared to control group(P<0.01); Compared to model group, the secretion and mRNA expression of TNF-α,IL-1β,IL-6 in catalpol group, berberine group and combination group decreased significantly (P<0.01); Compared to catalpol group and berberine group, the secretion and mRNA expression of TNF-α,IL-1β,IL-6 in combination group decreased significantly (P<0.01); Besides, there was no significant difference between the effect of catalpol group and berberine group on the secretion and mRNA expression of IL-6(P>0.05). The secretion and mRNA expression of TNF-α,IL-1βberberine group decreased significantly compared to catalpol group (P<0.01). Conclusion1. The consumption of glucose in IR 3T3-L1 adipocytes can be promoted by both catalpol and berberine, which indicates the two Chinese herbal drugs might be able to improve insulin resistance. In addition, the effect of the combination group is better than the single drug groups, showing the scientific connotation of Rhizoma Coptidis Pill.2. The secretion and mRNA expression of TNF-α,IL-1β,IL-6 decrease significantly by Catapol,Berberine and the combination of the Catapol and berberine . While the effect of berberine on the expression and secretion of TNF-α,IL-1βis more significant than that of Catapol, and there is no significant difference between the effect of Catapol on the expression and secretion of IL-6 and that of berberine . And compared with single drug groups, the combination group has significant effect on the secretion and mRNA expression of TNF-α,IL-1β,IL-6, suggesting that compatibility has synergistic effect.
