The Expression And Clinical Role Of Timp-2 In Hepatocellular Carcinoma In Hui Nationality

Objective Explore the expression of tissue inhibitors of metalloproteinase-2 in hepatocellular carcinoma and investigate the biological activities of this novel cytokine in the progress of the hepatocellular carcinoma in order to provide a basis and reference materials of gene diagnosis and treatment in hepatocellular carcinoma in Hui nationality.Method The expression of TIMP-2 was determined by S-P imunohistochemistry in 60 cases of Hui nationality and 18 cases of Han nationality in cancer tissues and 10 cases in healthy control tissues of Hui nationality.Result TIMP-2 was evaluated on several sections of HCC primary nodules and normal control tissues using polyclonal antibody. In HCC primary nodules, it was expressed in almost half of the samples examined, although some differences in terms of staining intensity were observed in the tissues of different patients. TIMP-2 staining was stronger in the primary nodule tissues of patients without metastasis than in those with. TIMP-2 was present mostly in the parenchyma, in the cytoplasm of cancer cells, but also in the extracellular space, In conclusion, TIMP-2 staining was inversely correlated with the proteolytic enzymes balance shifted in favor of ECM degradation.In peritumoral tissues TIMP-2 was mainly expressed in the cytoplasm of hepatocytes. In healthy control tissue, it was present scarcely, mainly in the cytoplasm of hepatocytes distributed in the parenchyma mainly around the centrolobular vein.The positive reaction (53.33%) in Hui nationality is slightly higher than in Han nationality (50.00%), but this was not statistically significance (Z=-0.258 P=0.796). Respectively there were no obvious difference in gender group (χ~2=0.375 P=0.540), age group (χ~2=0.950 P=0.330), AFP group (χ~2=1.455 P=0.228) and HBsAg group(χ~2=0.232 P=0.630). But there were significant differences in tissue group (χ~2=5.442 P=0.020), Edmondson grades group (χ~2=4.133 P=0.042), envelope group (χ~2=6.461 P=0.042), metastasis group (χ~2=4.275 P=0.039) and survival group(χ~2= 4.077 P= 0.043).Conclusion The level of TIMP-2 expression closely correlate with the occurrence, development and prognosis of HCC. TIMP-2 expressed similarly between the Hui and Han Nationality group, and are in substantial agreement with domestic and international research at present, which may be used as a prognostic marker for gene diagnosis and gene therapy in metastatic patients with HCC.
