| Hyperprolactinemia (HPRL) are a group of syndrome caused by many reasons resulted in reducing of prolactin (PRL) levels. The clinical performance contained galactorrhea, amenorrhea, menstrual scarce, fat, infertility, anovulation[1]. Now the main treatment drugs are ergot alkaloids, such as bromocriptine. However such drugs have many side effects and are more expensive, so patients have the poor compliance. Malt is a kind of Chinese medicine commonly use in clinical, have the effects such as digestion, appetizer, swelling and so on. The compound Chinese medicines are commonly used in treating HPRL, and the pre-clinical observation of single malt has show a satisfactory effect in treating this disease. So we make further studies on the treatment of malt in HPRL to confirm the active site and active constituent in treating this disease. And then develop the traditional Chinese drugs into a new Chinese drugs pharmaceutics that have exact effect and low-price, change the situation of depend on bromocriptine that is the major expensive importing drug to treat HPRL.The effect of total malt extract on PRL level in the female HPRL mice:Inducing the female HPRL mice model by metoclopramide, that is dopamine receptor antagonist drugs[2], using bromocriptine as the control, giving the high, medium and low-dose malt extract treatment, detecting the blood serum PRL of by radioimmunoassay, analyzing its results with statistical software. The results showed the blood serum PRL of the high, medium and low-dose total malt extract group were significantly decreased, the difference was significant (P<0.05), the total malt extract could inhibit HPRL induced by metoclopramide, the effect is similar to the role of bromocriptine. And determining the polysaccharide content of malt extract by the phenol-sulfuric acid method, determining the flavonoids content of malt extract by UV spectrophotometer.The effect of basic constituent malt extract on PRL levels in the female HPRL mice:The method is same as the part of 1. The results showed the blood serum PRL of the high, medium and low-dose basic constituents malt extract group were significantly decreased, the difference was significant (P<0.05). it is evidenced that the active constituent of inhibiting HPRL may be the basic constituent malt extract.Piper wallichii was a perennial herb of the genus piper, which belong to the family piperaceae. This plant was used for promoting blood circulation, alleviating pain, improving sexual function and the treatment of beriberoid disease[3]. In this study, the phamacognostic methods were applied, the extract of Piper wallichii was examined, moisture and ash were detemined, and the bioactive constituents of piperlonguminine were analyzed by TLC and HPLC. The established method can be used for the quality control of Piper wallichii. To provide scientific basis for the utilization and development of this herb and lay the foundation in developing new drugs.In our study, aristololactams, including Cepharanone B, Aristolactam Aâ…¡and Aristololatams Aâ…¢a, are seperated for the first time from piperaceae and have the renal toxicity[4]. So it is essential to determine the content of these constituents. At equal condition, the contents of three aristololactams were simultaneously determined in Piper wallichii by HPLC-UV, which could fully help to improve the quality control of this herb and would provide an important reference to establish the quality control method for other related traditional Chinese medicines preparations. Furthermore, the established method laid the substance foundation for toxicity studies. |
