ObjectiveGiant cell tumor of bone(GCT)is one of the primary bone tumors Which have strong invasiveness,poor clinical therapeutic response,sarcomatous change after radiation therapy,high recurrence rate and the tendency of pulmonary metastasis.Pathologic grading can't predict the tumor's biologic behavior correctly,which bring problems,such as correct diagosis and treatment.So it is important to identify a tumor specific maker which is important in the design of an interventional procedure,guiding for clinical decisions and judging prognosis. Therefore,this experiment to study the roles of CD 147 and Survivin in develop-ment and progression of bone tumors.To study the expression levels of CD 147 and Survivin in giant cell tumor of bone and detect the correlation between their expression levels and the histological grade,radiological stage,recurrence state of the tumor.To intend to find possible prognostic indications of the aggressive behavior of the underlying pathology.Materials and methodsThe clinical manifestations and changes of 34 cases giant cell tumor of bone(GCT)(Jaffe grade:10 case of JaffeⅠgrade,20 case of JaffeⅡgrade,4case of JaffeⅢgrade; Campanicci's grade:10 case of Campanicci's I grade,15 case of Campanicci'sⅡgrade,10 case of Campanicci'sⅢgrade) patients which were surgically treated by local excision in the Orthopeadic Department of our hospital from January,January,2000 to March,2009 was retrospectively reviewed. The expression of CD 147 and Survivin in 34 cases of giant cell tumor of bone was studied by using S-P immunohisto-chemical method, and compare to 10 cases osteosarcoma and 10 cases osteochondroma,to analyze the relationship between the expression levels and clinical-pathological features Jaffe grading, Campanacci's grading and the recurrence. ResultsThe level of the expression of CD147 and Survivin in giant cell tumor of bone was 44.12%(15/34)and 58.82%(20/34)respectively.There were no relevance between the expression of CD 147 and gender,age,invasive location,Jaffe grade and Campanicci's grade (P>0.05).but the expression of CD147 was relevant to prognosis(P<0.05).Expression of CD 147 was significantly higher in giant cell tumor of bone with recurrence than that in giant cell tumor of bone without recurrence.There were no relevance between the expression of Survivin and gender,age,invasive location,Jaffe grade and Campanicci's grade (P>0.05).but the expression of Survivin was relevant to prognosis(P<0.05).Expression of Survivin was significantly higher in giant cell tumor of bone with recurrence than that in giant cell tumor of bone without recurrence.Conclusions1.In different grade tissue of GCT,CD147 and Survivin proteins had the positive expression of the different degree.There was significant correlation between expression of CD 147 and Survivin and prognosis.The occurrence that expresses that the CD147 and Survivin may participate GCT evolve,both of expression appearance may affect the malignant degree and the prognosis progressions,which can be used as the new marking thing of tumor to diagnose GCT qualitatively.2.The CD 147 and Survivin proteins had higher expression and expressions had a agreement in GCT tissue. The positive expression of CD 147 and Survivin had nothing to do with gender, age, invasive location,Jaffe grade and Campani-cci's grade.but the expression of Survivin was relevant to prognosis (P<0.05). Expression of Survivin was significantly higher in giant cell tumor of bone with relapse than that in giant cell tumor of bone without relapse. CD147 and Survivin play a role in the occurrence and develop course of GCT3.The expression of CD147 and Survivin in osteosarcoma is obviously higher than that of osteochondroma,and the difference is significant.The expression of CD 147 and Survivin in GCT is also obviously higher than that of osteochondroma,and the difference is significant.The difference of CD 147 and Survivin expression between osteosarcoma and GCT isn't significant.CD 147 and Survivin can be a important parameter for determining malignant level and estimating prognosis in bone tumors. 4.Overexpression of CD 147 and Survivin in patients with giant cell tumor of bone can enhance the possibility of relapse implicating a poor prognosis.They may serve as the fairly good prognostic factors to indicate biologic behavior and prognosis in giant cell tumor of bone and the possibility of new therapy in giant cell tumor of bone.
