Objective:To observe the effect of KG ointment on xylene-induced mouse ear acute inflammation and DNFB-induced delayed-type hypersensitivity model mouse; to study its efficacy and possible mechanism in the treatment of acute eczema.Methods:1.60 KM male mice,18-22g, randomly divided into 6 groups according to the weight with 10 mice each group. The 6 groups are control group, matrix control group, positive control group (dexamethasone), and high, medium and low dose group. The mice are treated with KG ointment for consecutive 4 days, once a day on the left ear during the previous three days; twice a day on both ears on the fourth day. One hour after the last administration, drop 20ul xylene in the mice's left ears, kill them one hour later by cervical, cut down the ears long the baseline position, weight ear variation, which is called swelling.2.60 KM male mice,18-22g, randomly divided into 6 groups:normal control group, model control group, KG ointment high-dose group, KG ointment dose, KG ointment low-dose group, positive group (dexamethasone)., A mouse delayed-type hypersensitivity model is established with the method of DNFB, abdominal sensitization arid baseline stimulation. After the mice are killed, their ears are cut off along the baseline position, weighed and calculated as swelling. Put the left ear in 10% formalin fixation, stain it with HE congestion and observe the degree of edema, hyperemia and inflammatory cell infiltration; observing the influence of KG ointment on TNF-a, IFN-y, IL-4, ICAM-1 levels in mice tissue with the tool of IHC.Results:1. Compared with model control group, mice ear swelling in high and middle dose groups is less than that in model control group, with statistical significance (P<0.05 or P<0.01); the low dose group shows no significant difference (p> 0.05), but some tendency of decrease. The result shows that KG ointment has inhibiting efficacy on xylene-induced ear swelling in mice and has anti- acute inflammatory effect. 2. Compared with model control group, each administered group is less than that in model control group, with statistical significance (P<0.01). The pathological grade of KG ointment in high, medium and low dose groups decrease obviously, with statistical significance (P<0.01). The results show that KG ointment can reduce the DNFB-induced mice ear swelling, can reduce the pathological damage in the left ear, and can inhibit delayed-type hypersensitivity in mice caused by DNFB.3. Comparing model group with normal control group, it is found that in the DNFB-induced mice ears, IFN-γ, IL-4, TNF-α, ICAM-1 levels are significantly higher, with statistical significance (P<0.05 or P<0.01); the percentage of IFN-γ, IL-4, TNF-α, ICAM-1 in each administered group is lower, with statistical significance (P <0.05 or P<0.01), The result shows that KG ointment can inhibit the expression of these cytokines.Conclusions:1. KG ointment can inhibit xylene-induced mouse ear inflammation and 2,4-dinitrofluorobenzene-induced delayed-type of hypersensitivity in mice, can improve the infiltration of ear tissue inflammatory cells, and reduce cell swelling, which indicates that KG ointment has efficacy in the treatment of acute eczema.2. The mechanism of KG ointment in treating acute eczema is related to the expression of IFN-γ, IL-4, TNF-α, ICAM-1 and the regulation of the function of T cell subsets.
