Objective:This study was designed to investigate a quality control method of ABO typing of umbilical cord blood (UCB) by using routing serologyContent:Compare serology positive typing (Serum-paper pieces) with reverse typing (Tube test) accuracy. From three parties, the human factor, reagent factor and the typing method to explain the necessity of the review. Review and discuss the various factors, which influence the results of ABO typing of umbilical cord blood. Introduce a very simple, fast and effective technique for the isolation of pure high molecular weight genomic DNA in whole blood. Those samples with questions were further defected by Sequence Specific Primer PCR (PCR-SSP). In the end, comparison with the result of genotyping, positive ABO typing and reverse typing.Methods:UCB samples identified by positive ABO typing. Those samples with questions were further defected by PCR-SSP.Results:Among total of 76120 UCB samples identified by positive ABO typing, there were 78 samples (1‰) could not be determined. Of these 78 samples,30 were included as a weak agglutination reaction by reverse ABO typing. Out of 260 samples in reverse ABO typing,112 cases (43.08%) were inconsistent with the positive ABO typing.58 undetermined samples were detected by PCR-SSP. The genotyping results of 45 samples were conform to serological typing.3 cases of inconsistent.10 with unconformity between positive and reverse typing, which genotyping results were fully consistent with the positive typing.Conclusion:Positive typing for red cell antigens combined with PCR-SSP is efficient and sensitive for quality control.
