Research purposes:In order to regulate acupuncture treatment, objective and sci-entific assessment of efficacy, intended use of basic theories of traditional Chinese medicine combined with modern medicine, research methods, the use of animal mo-dels of hypoxia-ischemia of the newborn, the use of molecular biology, imm-unology technique to observe the time under different acupuncture the added value of brain st-em cells, differentiation, and explore the acupuncture treatment of neonatal hypoxic-ischemic encephalopathy (hypoxic-ischemic encephalopathy, HIE) mechanism, clear-ly the best acupuncture treatment of this disease limitation. Acupuncture treatment for HIE to provide scientific, solid theoretical basis for the comprehensive treatment of HIE to find more suitable for the characteristics of the safety of children, practical and effective way.Materials and Methods:The health of newborn male and female rats in any seven days, weight 12g~14g, were randomly divided into normal (blank) control group 24, model 24, rapid gill no acupuncture group (gill-speed) 24, leaving 24-pin 30min group, After three groups of left common carotid artery ligation and hypoxia for 2 hours produced the newborn rat model of hypoxic ischemic encephalopathy. Each group is divided into 3 days,7 days group,14 days and 28 days groups, respectively, after the first 3 days,7 days, the first 14 days and 28 days for materials; at all obser-vation points at the same time the four groups drawn. Four groups of the same feeding conditions.Acupuncture Method:Controlling Vessel,Governing Vessel, given fast no thorn needle, leaving the needle 30min, once a day. Normal control group and model control group not dealing with acupuncture. Animals in each group based on two hours before the intraperitoneal injection of Brdu (5-bromodeoxyuridine, BrdU) for the tag brain neural stem cells proliferation. Model, respectively, after 3d,7d,14d and 28d, the use of neurological deficit score and HE staining of the nerve function of animals with symptoms of the pathological changes of brain injury; immuneohistochemical observation of hypoxic-ischemic side of rat dentate gyrus (DG), subventricular zone (SVZ) of 5-bromo-deoxy uridine (Brdu), nestin (Nestin) changes, after the detection of cerebral ischemia in the number of neural stem cells and their proliferation, and thus speed rating gill stay for the hypoxic-ischemic brain injury the role of neural stem cells.Results:1.speed thorn, leaving for the HIE rats with neurological deficit scores of the impact:The Kruskal-Walhs test (normal control group, excluding one), all operated rats 28 days after the symptoms of neurological function significantly reduced compared with day 1 (P<0.01). In each pairwise comparison between groups, the speed group and stay in stab-acupuncture group of neurological symptoms in rats after 14,17,21,28 days than in model group was significantly reduced compared with the model group were significantly different (P<0.05).2. speed thorn and stay for the HIE in rat brain tissue morphology of impact:model control group:left-brain hippocampus shows focal necrosis, coagulation necrosis region was reduced nucleus-like, necrotic foci surrounding normal level of a clear organizational structure; left brain occipital cortex shows the organizational structure of fuzzy large necrotic areas; speed gill treatment groups:the majority of nerve cell degeneration and a small amount of nerve cell degeneration and necrosis; stay acupuncture group:the majority of nerve cells is still good, a small amount of nerve cell degeneration and necrosis.3.speed model of thorns and stay for the HIE protein expression in rat brain tissue BrdU effects:with the normal control group, model control group rat brain DG and SVZ zones BrdU protein expression increased significantly (P<0.01); speed thorns and needle can be left to BrdU protein upregulation. With the model control group, treatment group and stay in stab-speed needle treatment group BrdU protein expression increased (P<0.01), with statistical significance. Speed thorn acupuncture treatment group and stay in group, after 3d,7d, when protein expression of BrdU had no significant difference (P> 0.05), after 14d,28dBrdU protein expression was significant difference (P<0.05), left-acupuncture group expression is higher than speed gill treatment group.4.speed thorns and stay for the HIE model Nestin protein expression in rat brain tissue effects:Compared with normal control group, model control group rat brain DG and SVZ areas Nestin expression increased significantly (P<0.01); and model the control group, treatment group and stay in stab-speed needle treatment group Nestin protein expression increased (P<0.01), with statistical significance. Speed thorn acupuncture treatment group and stay in group, after 3d,7d, when the expression of Nestin protein was no significant difference (P> 0.05), after 14d,28dNestin protein expression was significant difference (P <0.05), left-acupuncture group expression is higher than speed gill treatment group.Conclusion:1,Speed thorn and retaining can promote recovery of limb function in HIE rats, lack of nerve function score had improved significantly.2,Speed thorn and retaining can HIE area in rat DG area and BrdU-positive SVZ cell proliferation and more pronounced, indicating that Speed thorn and retaining so that the increased proliferative activity of the two regions.3,Speed thorn and retaining can HIE in rat DG area and SVZ area Nestin-positive cell proliferation more evident, indicating that barbed-speed and stay in acupuncture for neural stem cell proliferation.4,Speed thorn and stay of acupuncture on HIE in rat DG area and SVz of neural stem cells in early stage non-discriminatory; section 14d and section 28d to remain superior speed needle stab.5,The experimental results showed that speed thorns and stay acupuncture can promote cerebral ischemia and hypoxia of neural stem cell proliferation, differentiation, promote nerve cell repair, to reduce functional impairment, which may be the important role of acupuncture is one of the mechanisms HIE.
