ObjectiveTo explore the genomic amplification and clinical significance of the human telomerase gene(hTERC) in cervical adenocarcinoma as well as the infection characteristics of HPV genotype,in order to ascertain the role of hTERC as an early biomarker of cervical adenocarcinoma and provide an theoretics basis for the screeing and prevention for the precancerous of cervical adenocarcinoma.MethodsThe study includes formalin fixed paraffin embedded tumour tissue specimens from 40 cervical adenocarcinomas, diagnosed and surgically treated at the Shandong University Qilu Hospital during 2007 to 2009 and the cervical cytologic sample of 10 of cervical adenocarcinoma。(1) All tumour cases were collected to study the amplification of the hTERC gene in cervical adenocarcinomas by using fluorescence in situ hybridization(FISH) with a duble colour probe set specific for centromeres of chromosome 3 and the TERC gene. (2) Extraction of DNA from formalin fixed paraffin embedded tumour tissue and to detect the geno typing of HPV by low density gene chiP.Results(1) in 40 samples,77.5% of those were checked out HPV and the major epidemic subtypes of HPV were HPV16(42.5%) and HPV 18 (27.5%).(2) The multiple infection rate of HPV in cervical adenocarcinomas is 7%. (3) We observed high proportions of nuclei with increased absolute copy numbers for TERC in all tumours (range 2.4-4.9). Expression rates are 100%. Conclusion(1) In this data, the major epidemic subtypes of HPV were HPV16(42.5%), HPV18 (27.5%), it proved preferential risk of HPV16 for squamous cell carcinoma and adenocarcinoma of the cervix and of HPV18 for adenocarcinoma of the cervix.(2) The multiple HPV infection is not an indicator of severity of cervical lesion. May be the severity of adenocarcinoma of the cervix did not rely on the quantity of the multiple infection but depend.on the virulence of HPV genetype.(3) Amplification of the human telomerase gene TERC is a consistent aberration in cervical adenocarcinomas. The amplification in gene hTERC was a useful auxiliary indicators for diagnosing and screening of cervical adenocarcin-oma and precancerous.
