Objective:To observe the effects of Recombinant human erythropoietin (rhEPO) on random flap survival rates in the Sprague-Dawley (SD) rats and to discuss possible mechanism.Methods:A dorsal,caudally based random flap measuring 2cm×9cm was created in all 24 SD rats which were divided into two groups:Control Group (Group A, n=12) with saline injected through the intraperitoneal route after operation and treated Group (Group B, n=12) with single-dose rhEPO 5000 IU/kg injected through the intraperitoneal route after operation. At 24th hour postoperatively, the tissue myeloperoxidase (MPO) activity and amount of malondialdehyde(MDA) were evaluated on the flap tissue samples of 6 rats chosen randomly. The flap viability and immunohistochemical (CD34) methods which reveal the microvascular network were evaluated on rest rats at day 7 after operationResults:In the MPO activity test, the mean result of treated group is 0.116±0.017U/g,and the mean result of control is 0.148±0.018U/g, compared to control group, a significant decrease in treated group was observed (p<0.05). Amount of MDA tests, the mean result of treated group is 1.07±0.27nmol/mg-protein ,and the mean result of control is 1.46±0.25nmol/mg-protein compared to control group, a significant decrease in treated group was observed (p<0.05). In the flap viability test, the mean viability ratio of treated group is 66.47%±4.08, and the mean viability ratio of control group is 56.37±7.13, a significant increase in treated group was observed (p<0.05). In the microvessel density by CD34 immunohistochemical staining sections methods observation, the mean result of treated group is 131.9±23.15 positive cells per microscope field vision, and the mean result of control is 98.3±18.36 positive cells per microscope field vision , and a significant increase in treated group was observed(p<0.05).Conclusion:rhEPO augment the viability of rats dorsal random flap may by reducing neutrophil infiltration of flap tissue. The hematopoietic and angiopoietin effects may related to improving compliance and skin color after flap survived.
