Abstract/Summary:
Establishment of Real-Time Fluorescence Quantitative PCR (FQ-PCR) for Escherichia, Staphylococcus, Lactobacillus and analysis quantity change rule of the microflora in enteron (the whole esophagus duodenum, jejunum, ileum, caecum, recrum ) and respiratory system for Cherry-valley duck immuned with Low Duck-Plague-Virus (DPV).1. FQ-PCR method of the three genus was established, The sensitivity of this assay, Escherichia's is 23.4 cells/μL, Staphylococcus' is 2.24 cells/μL, Lactobacillus' is 6.98 cells/μL. Escherichia, Staphylococcus and Lactobacillus can be specifically distinguished from others flora and quantificationally detected with the FQ-PCR method .2. Microflora variety of Cherry-valley duck immuned by-mouth with low DPV and cohabitation duck in enteron and respiratory system.(1) Escherichia flora of esophagus trachea quantitative range (logcop/whole) <6.31-11.09, 7.58~>11.31, feces' quantitative range (logcop/g) <6.31 ~ 10.83. The result compare with control duck's, show wave variety and quantity rise. The distributing quantity is more in back intestines.(2) Staphylococcus flora of esophagus , trachea quantitative range (logcop/whole) 0.00—11.01, 0.00 ~ 11.28, feces' quantitative range (logcop/g) 0.00 ~ 11.16. The result compare with control duck's, the difference is not evident.(3) Lactobacillus flora of esophagus, trachea quantitative range (logcop/whole) 0.00~11.09, 0.00~11.09, feces's quantitative range (logcop/g) 0.00~ 10.61. The result compare with control duck's, the difference is not evident. Lactobacillus flora quantity of ileum, caecum , recrum rise along with old day. This is different to control duck's.3.
