| Combining the technique of multiplex-PCR and the fluorescent automated diction, the polymorphism of microsatellite was performed in new strains of BM and BF in silkies. In addition , the linkage correlation between microsatelite markers and characters of the age at first egg> the egg production of 300 days. the egg production of a week et al was also analyzed . 18 pairs of microsatellite primers were screened from chromosome 1 and chromosome Z, the primers were designed to allow optimal performance of the markers, in sets containing multiple microsatellites, on ABI3100 sequencers. We determined there were correlation among 13 microsatellite markers and 19 characters in 25 egg traits through least square means. At the same time, we calculated correlation among characters of egg traits in silkies. We can draw some conclusion as blow:1) Through multiplex-PCR reaction system ,we succeed in combining multiplex-PCR primers to their optimal performance;2) The 13 microsatellite markers loci which linked with egg traits accorded with the characters of Mendelian co-inheritance, the heterozygosities ranged from 0.198 to 0.775, the means of heterozygosity is 0.439, with 23% of loci having a heterozygosities of> 0.60; the polymorphism information contend (PIC) ranged from 0.408 to O.Sfcl, in which 84. 6% of loci had a PIC of more than 0.50, the means of PIC is 0.698; the average number of alleles for the 13 polymorphic markers described was 8.54, this indicated microsatellite markers can be used in linkage analyze ideally;3) Trough least square means, 13 microsatellite markers and 19 characters of egg traits were detected, which were correlation or correlation significantly, this indicated: The microsatellite markers described are particularly well suited for performing QTL andMAS;4) Difference among parent and FI is significant in the age at first egg and the egg production of 300 days, the heterozygosity and variation factor decreased , this indicated that the selection effect is significant through one generation selection;5) There were significantly negative phenotypic correlation between the age at first egg and the egg production of 300 days; A positive phenotypic correlation was found among theegg production of 300 days and the egg production in every week. A negative phenotypic correlation was found between the age at first egg and the accumulated egg production of 18~21week. |
