In BM and BF new layer breeds of silkies, the relationship between microsatellite DNA markers and egg quality traits was studied using multiplex PCR and fluorescence based full-automated electrophoresis. Linkage analysis between marker genotypes was studied with Duncan's Multiple Range Test in SAS software. In order to take advantage of multiplex PCR, the conditions was optimized individually and in combination. The results as follows:(1) On the basis of this optimization experiments, 18 microsatellite DNA markers were classified into 5 different sets. And then, the outputs of multuplex PCR were classified into other 3 sets, and detected on ABI 3100-Avant Genetic Analyzer. The results suggested that the optimization test of multiplex PCR successfully.(2) 18 pairs of microsatellite DNA markers were polymorphic. The average number of alleles detected by a pair of primers was 7.444. The average value of PIC, average observation population heterzygosity and expection population heterzygosity of the 18 markers were 0.669, 0.3962, and 0.7301, respectively.(3) Data analysis was performed with SAS6.12. We found there were correlation among 8 markers and 8 egg quality traits.The results of Duncan's Multiple Range Test shows: (1)Means of shell strength and shell proportion for the genotype II of MCW0289E, means of shell strength and shell proportion for the genotype AG of MCW0258, and means of shell strength and shell proportion for the genotype CC of MCW0068 were significantly higher than those of others. The result implied that genotype II of MCW0289E, genotype AG of marker MCW0258, and genotype CC of MCW0068 may be regarded as candidate locus for early assistant selection markers in shell strength and shell proportion traits of silkies. (2)Means of yolk proportion for genotype HH of MCW0289E was significantly lower than those of others. Means of haugh unit for genotype HH of MCW0289E was significantly higher than those of others. Means of yolk proportion for genotype JO of MCW0258 was significantly higher than those of others. Means of haugh unit for genotype JO of MCW0258 was significantly lower than those of others. The result implied that genotype HH of MCW0289E and genotype JO of MCW0258may be regarded as candidate locus for early assistant selection markers in yolk proportion and haugh unit traits of silkies.(3)Means of egg shape index for genotype BG of MCW0289E was significantly higher than those of others. Means of haugh unit for genotype BG of MCW0294 was significantly lower than those of others. Means of egg shape index for genotype JL of MCW0258 was significantly lower than those of others. Means of haugh unit for genotype JL of MCW0258 was significantly higher than those of others. The result implied that genotype BG of MCW0294 and genotype JL of MCW0258 may be regarded as candidate locus for early assistant selection markers in egg shape index and haugh unit traits of silkies.At the same time, we found some alleles associated with egg quality traits: (1) Means of shell strength for genotype AG and BG of MCW0258 were significantly higher than those of HO and JO. The allele G and O of MCW0258 had a significantly positive and negative correlation with shell strength respectively. (2)Means of egg shape index for genotype JL and JJ, LL, FL of MCW0258 were significantly lower than those of others. The allele J and L of MCW0258 had a significantly negative correlation with egg shape index. (3)Means of shell strength for genotype CC of MCW0068 were significantly higher than those of others. But means of shell strength for genotype CI and BI of MCW0068 were significantly lower than those of others. The allele I of MCW0068 had a significantly negative correlation with shell strength.(4) There were significantly correlation among the 9 egg quality traits in silkies. There were positive phenotypic correlation among albumen proportion and egg weight (P<0.01), Haugh Unit and albumen proportion (P<0.05), shell proportion, shell strength and shell thickness (P<0.01) respectively.
