| Compared with the traditional technology of extraction and separation, flocculation technology shows special superiority such as reduce the production cycle,lower the energy consumption,ensure the retention rate of available components as well as increase the clarity and stability of the pharmaceutical product. In this paper,flocculation technology was applied to extract and separate polysaccharide from jujube at the first time.Chitosan and ZTC1+1Ⅱas the flocculant used in the process of extracting and separating polysaccharide from jujube was researched.Based on these studies two new extraction processes of polysaccharide were found.At same time,high-performance liquid chromatographic method for checking cyclic Adenosine-3',5'-monophosphate(cAMP)was investigated in this article.HPLC was applied in order to analyse cAMP in jujube from Xinjing and Shanbei and make a comparison between them.Firstly,the technics flow of flocculation technology was confirmed according lots of references.And the analysis was also ascertained.The transparence and colour of the liquid was determined by spectrophotometry.Phenol-sulfuric acid was used for the analysis of polysaccharide,while Coomassie brilliant blue G-250 was applied for the analysis of protein.Secondly,chitosan was applied to extract and separate the polysaccharide from jujube.The clarification effects of the chitosan's dosage and molecular weight,the reaction time,temperature and pH were investigated.Then,by making the content of polysaccharide from the jujube as index,the best reaction condition was found according to the result of orthogonal test.The chitosan best dosage was 3mg/ml, While the molecular weight was 100 thousand.The reaction only needed about 2 hours.And the temperature should be controlled at 30℃.Thirdly,ZTC1+1Ⅱwas applied to the extract and separat polysaccharide from jujube,too.ZTC1+1Ⅱcontains two components about A and B.After the charging sequence of two component,the clarification effects such as the dosage of ZTC1+1Ⅱ,the reaction temperature and pH were investigated.According to the research,the charging sequence of two component was ascertained.In order to having a good effect, B component should be added firstly.Then,by making the content of polysaccharide from the jujube as index,the best reaction condition was found according to the result of orthogonal test.The dosage of ZTC1+1ⅡB was 0.70mg/mL,while the other was 0.350mg/mL.The best reaction temperature and time was 30℃and 1 hour.Lastly,high-performance liquid chromatographic method for checking cAMP also was investigated in this article.The optimum parameters of HPLC were determined to detect cAMP,which was extracted under certain conditions by using different methods.Agilent XDB-C18(4.6mm×150mm,5μm)column was used as the solid phase while the mobile phase was methanol-0.05mol/L KH2PO4(10∶90).The flow rate was 1.0mL/min.The content of cAMP was measured by Diode Array detector under 259nm.And the temperature of the detector was 35℃.The results showed that the condition of HPLC in this article could separate cAMP from other compositions in jujube.There was different content of cAMP in jujubes from different areas.According to the quantificational analytical results,the content of the cAMP in the jujube from Xinjiang was 320μg/g and the other from Shanbei was 306μg/g.And this investigation showed that this method was sensitive,rapid and accurate. |
