Rodents have played important role in maintaining the structure and function ofbiocoenose and the flow of material and energy among ecosystem.However,in recent years,rodents have turned to be more and more dangerous for most habitats.Cricetulus barabensis,as the dominant rodent species at North of China has caused more and more attention for its high reproductive capacity.FSHβgene was considered as a related gene for the reproductive capacity of animals.1 198bp of FSHβgene was cloned using PCR technique in the species of C barabensis in Shandong Province for the first time.The primers were designed according to the homologous species from GenBank This part of FSHβgene was then cloned and sequenced in each 30 individuals in Yinan,Linqu and Qufu in Shandong Province.A total of 13 SNPs were examined, including 4indels Insert/Deletion and the ratio of conversion to transversion was 8.13 haplotypes were defined in 90 samples.The sequence numbers submitted to the GenBank were FJ882151~FJ882163.The sequence of exon 1 and exon 2 of FSHβgene obtained from C barabensis were compared with the corresponding sequences of the other 20 species embodied in GenBank.It showed high identities in the level of both nucleotide acids(82%100%) and amino acids(70%~100%).The nucleotide diversity(π),haplotype diversity(Hd),number of haplotye(H),number of polymorphic sites(S),average number of nucleotide differences(K) and theta from S(θw) of FSHβgene of Y'man,Linqu and Qufu populations were calculated with DnaSPv4.10 sol,ware,which showed that the genetic diversity of the three populations arrayed as Yinan,Qufu and Linqu.In addition,Fst,Nm and Dxy proved the genetic differentiation among the three populations existed in a low level.The result of Gst analysis among three populations arrayed from 0.01 to 0.05,which meant that 95%~99%mutation existed in each population while 1%~5%mutation existed among populations.NJ analysis showed that 90 samples intersected with each other instead of forming three different groups consistent with geographic differentiation,which demonstrated the genetic differentiation among the three populations was not distinct.The research showed that FSHβgene on C barabensis from Yinan, Linqu and Qufu was rich in haplotype diversity and nucleotide diversity.It needs further research to fred the relationship between the FSHβgene and the high productive character of C barabensis.Partial small-insert genomic library composed of more than 3 000 clones was screened with PCR technique for recombinant positive clones containing microsatellite sequences of C barabensis.A total of 18 microsateUites were isolated.The sequence numbers submitted to the GenBank were EF543177~EF543190,EF489000,EF489001.Among the 18 microsatellites. perfect ones were 11,accounted for 61.1%;imperfect ones were 4,accounted for 22.2%; compound pertect ones were 3,accounted tor 16.7%.High homologous microsatellites had not been found in GenBank,which indicated that the 18 microsatellites were isolated for the first time.These 18 microsateUites further enriched the molecular markers of C.barabensis.
