Partial small-insert genomic library composed of more than 3000 clones was screened with PCR technique for recombinant positive clones containing mierosatellite sequences of Cricetulus barabensis.The primers used in PCR were M13 Primers M3/M13 Primers RV which located at both sides of multiple clone site of pUC19 and microsateUite primers (CA)8/(GA)8designed according to core repeats motifs of microsatellite.About 69 recombinant positive clones containing microsatellite sequences of Cricetulus barabensis were obtained through rapid screening small size fractionated genomic libraries with PCR technique.Sequencing of these clones was performed with automated sequencer,and 36 microsatellites were isolated.The sequence numbers submitted to the GenBank are EF543177~EF543179,EF543184~EF543187,EF489000,EF489001,EF491717,EF530600, EF530605~EF530607,EF530609~EF530611,EF530613,EF530615,EF530618,EF530621, EF530623~EF530625,EF502036,EF532795.The motif number from 8 to 20 in most microsatellites are 14 accounted for 39.9%,while the motif number from 21 to 38 are 8, accounted for 22.2%.In the 25 microsatellites,perfect ones are 21,accounted for 57.3%; imperfect 28.8%,compound perfect 13.9%Ten microsatellite sequences were selected for desiring oligonucleotide primers,using Lasergene 6 and Primer 5.0 software.The primers that have some multiplicity character were screened out by the agars gel-electrophoresis and polyacrylamide gel-electrophoresis so as to detect the multiplicity locations of Cricetulus barabensis living in different regions.At the same time did some statistical analysis.The result indicated that six microsatellite markers were polymorphism.Genetic polymorphism of Cricetulus barabensis populations in Qufu Wucun,Neimeng Shadi and Neimeng Yangcao were studied with these primers.The result indicated that genetie variation of Qufu Wucun population was highest and the lowest population was Neimeng Yangcao.Cluster analysis showed that there were differences between the Qufu Wucun population and Neimeng Shadi and Neimeng Yangcao.
