Since rich in nutrition, Spirulina is widely used in food, cosmetic, medicine and feedstuff areas. And it has a brighter future to be a producer in ecological guarantee system. The photoautotrophic and mixotrophic culture of Splatensis were studied in this paper, which was focused on the the culture characteristics of Spirulina and combined with the studies of our previous research. And the effect of microgravity condition on the growth of Spirulina was studied from growth status, the aspects of content of major compounds, and so on.The phototrophy and mixotrophy culture conditions of Splatensis in shaking flask, stirred tank fermenter and self-made airlift reactor was optimized. The culture conditions of Splatensis in elf-made airlift reactor and the content of major compounds and the causes of difference in major compounds were also studied. The best culture conditions were: rotational speed 11rad/min, light intensity 3.5klx, culture time 10d, glucose concentration 2.0g/L, and Na2S2O3 concentration 1.0g/L. The maximum biomass of 0.98g/L, polysaccharide 33.78%, phycobiliprotein 36.50% and chlorophyll 45.79mg/g were obtained under phototrophy culture. And the maximum biomass of 1.35g/L, polysaccharide 24.55%, phycobiliprotein 72.72% and chlorophyll 33.80mg/g were obtained under the best conditions. After compared the results, it showed that microgravity condition will affect the composition content significantly, especially the strongest on phycobiliprotein. And it was 1.811 times than mixotrophy culture in shaking flask.
