Comparative studies between Ad-6 and ZGC (Oiychophragnnis violacetts ) onhypocotyl growth, bourgeon of cotyledon axillary bud, shoot culture, callus inductionand culture of unpollinated ovary and ovule were conducted. Ad-6, with restorer genesfor Ogu CMS, is the progeny of cross Raphcimis sativm L var. RaphanislmidesMakino (♂)× Brassica napus (♀).Cytokinin can moderate growth of hypocotyl which shows abrupt growth in the absence of plant growth regulators, and the effect of cytokinin on Ad-6 is more conspicuous than on ZGC. Cotyledon axillary buds of Ad-6 is more sensitive to 6-BA than that of ZGC. 0.5 mg/16-BA can induce bourgeon of Ad-6 cotyledon axillary buds, with no effect on ZGC. 0.1-0.5 ing/1 NAA can make shoots of Ad-6 and ZGC to produce lots of short and strong roots which can assure successful transplant in the soil. Addition of a little NAA alone in MS medium is helpful to the prolonged storage of shoots without vitrification. ZGC shoots have higher tolerance to 2,4-D, which is also a kind of herbicide, than Ad-6 shoots. GA, is important for multiplication of ZGC shoots. 0.1 mg/I GA, can lengthen stern internode of multiplied ZGC shoots, but less effect on Ad-6 shoots. It is easy to induce calluses of Ad-6 sections. But for ZGC sections, difficult to get calluses and easy to get regenerants. High dosage of 2,4-D (0.5 mg/1 or above), which can inhibit regenerants formation of ZGC sections, is important for ZGC callus induction.For culture of unpollinated ovary and ovule, flower buds with pedicel were firstlycultured to get enlarged ovaries (anther will not dehisce under high humidity), then ovules were excised for callus induction from enlarged ovaries in 7 days. Flower appendages (petal, sepal and pedicel), 6-BA and erect placement of flower buds are helpful to ovary enlargement. Sucrose, Macronutrients, and Vitamins of MS medium are more important for ovary enlargement than Iron and Micronutrients. Flower buds collected in cool season are better for induction than that in hot season, but 4℃ precondition of in vitro flower buds is harmful to ovary enlargement. Flower buds that will flower in 1-2 days are better for culture than smaller buds. Ad-6 ovaries is more sensitive to 6-BA than ZGC ovaries. Auxin is indispensable for ovule callus induction, accompanying with the promotion of 6-BA and GA3, Ad-6 ovule calluses regenerated only roots. But ZGC ovule calluses regenerated many shoots, among which haploid plant was found.
