With the development of researches on the mechanism of NO,NOS and ET action in animal economy, more and more attentions are paid to their function during the period of gestation. Nowadays, researches on the effect of NO,NOS and ET on gestation mainly focused on the change of the level of NO and ET in blood plasma,the orientation and distribution of NOS in uterus as well as the immanence connection between NO,NOS,ET and some disease during the period of gestation such as pregnancy-induced hypertension syndrome(PIH),endometrium cancer,pre-eclampsia and so on. However, until now, we found no research on the change of NOS positive nerve cells in spinal cord and the change of the content of NO and ET in pallium during gestation. The purpose of this paper is to discuss the effect of NO and ET on the process of gestation by analyzing the change of NOS positive nerve cells in spinal cord and the content of NO and ET in pallium in the metaphase of gestation, to provide gist on the understanding of mechanism of NO and ET action in nervous system on gestation, and to provide base data for examining the reason of gestation failing in clinic medicine and for assuring the normal growth of the fetal.In this study, mice at the metaphase of gestation were taken as study object and the control group was non-pregnant ones.We used the method of NADPH-d to show the conformation and distribution of the NOS positive nerve cells in spinal cord at the sect of neck,chest and waist of mice at the metaphase of pregnancy, and used Motic images Advanced 3.2 to analyze the area of the NOS positive nerve cells at these parts. We mensurated the content of NO in pallium of mice in the metaphase of pregnancy and of mice that were non-pregnant comparatively with the method of nitric acid reductase, and using radioimmunological method to analyze the content of ET in pallium of mice in the metaphase of pregnancy and of mice that were non-pregnant comparatively. The statistical analysis was carried out by SPSS 13.0, and the significance of difference was tested by t-test and one-way ANOVA.The results showed as follows: (1)there was significant difference on the area of NOS positive nerve cells in spinal cord at the section of neck and waist between mice in the metaphase of pregnancy and mice that were non-pregnant (P<0.01), and the difference of NOS positive nerve cell in spinal cord between them at the section of chest was also at the significant level(P<0.05); (2) there was no significant difference on the area of NOS positive nerve cells in spinal cord between the section of neck and waist of mice in the metaphase of pregnancy (P>0.05), but the difference between the section of neck and waist, chest and waist was at the significant level(P<0.01). (3) There was significant difference on the content of NO in pallium between mice in the metaphase of pregnancy and mice that were non-pregnant (P<0.01); (4) the difference on the content of ET in pallium between mice in the metaphase of pregnancy and mice that were non-pregnant was also at the significant level (P<0.01).We get these conclusions from the results: the area of the NOS positive nerve cells in spinal cord at the section of neck,chest and waist of mice in the metaphase of pregnancy are larger than that in the non-pregnant mice of the same section. This change may be propitious to increase the permeability of vein and provide a well condition for the development of embryo. The area of the NOS positive nerve cells in spinal cord at the section of waist which is conjoint with uterus are far larger than that at the section of neck and chest, and this difference may demonstrate that the waist section may play an important and direct role on the modulation of physiological action during gestation.The content of NO which can stretch vein reduced evidently in pallium during gestation while the content of ET which can shrink vein increased apparently at the same time. These changes enhance the blood pressure and speed the blood stream, so it could increase the matter exchange between matrix and fetal and offer enough nutrition for the normal development of fetal. There was close relationship between the nervous system which pallium belongs to and the course of gestation, and it control the complicated physiological course indirectly.The dilating and shrinkage of the vein during the stage of gestation could maintain the balance of blood pressure in vivo and provide a well environment for the development of the embryo. The change of the content of NO and ET in pallium and the increase of the area of the NOS positive nerve cells in spinal cord of mice in metaphase pregnancy might provide theoretical basis for explaining the action which central nervous system exerted through NO,NOS and ET during the stage of gestation, as well as provide theoretical evidence for further probing into the methods for resolving PIH in clinic.
