Celery is a kind of medicinal and edible plants,there are a lot ofbioactive materials contained in celexy,such as celery flavonoid,unsaturated fatty acid,essential oil, coumarin derivatives and so on.Among them,pelargidenon,persicarin,luteolin are promising in antioxidation,curing down lipid peroxidation,prevention of cardiovascular disease and anti-aging.But these substances have not been effectively utilized until now. As celexy is of low price and high yield,we selected as our research material and studied the techniques to separate and purify the total flavonoid from them.In this assay,the effect of extraction solvent on extracting the flavonoid was inspected.Moreover on bases of this,with orthogonal experiments,the optimal condition are obtained as follows:the concentration of alcohol 80%,solid liquid ratio 1:30,tempercture 70℃,extraction time 120 min.AB-8 macroporous adsorbing resin was adopted after comparison with other materials on separation and purification abilities.Crucial parameters like injection pH,injection rate,eluent concentration,elution time and elution speed were all optimized through screening,then we obtained the total flavonoid purity of crude extract 5.83%.We deterimend the most optimal techniques of extraction,separation and purification of total flavonoid from celery,and diastase was utilized in the step of purification for the first time.The results showed the product purity was greatly enhanced and could be as high as 34.68%.The antioxidative activity of product was not destroyed when tested through antioxidation experiment,which testified the feasibility of using diastase in purification of celery flavonoid.
