Auxins are important phytohormones, which are concerned with plants' growth and development, and they play importmant roles in plants' growth. There is little research about kelp auxin abroad and home, quantitative and qualitative analysis is still at the underway phase and there is no moulding technology, there are only a few reports about content of individual seaweed auxins but little reports on studies about kelp auxin. This article establishes the technique of extraction and separation of auxin in kelp through systemic experiments, and the kelp auxin act on several marine micro-algaes' growth with a distinct effect.On the basis of extraction of terricolous bioactive substances, we did orthogonal experimental design to lots of factors, and optimized the conditions of isolation and purification from kelp. In this paper, the process was established including tissue slurry at 4℃, lixiviation with 95% ethanol, extraction with ether, decoloration with silica gel, purification by Sephadex LH20, quantitative and qualitative detection and estimation by RP-HPLC. The process lays a foundation for further study on bioactivities, exploiture of plant growth accelerants and other regulators in kelp with convenience, quickness, accurateness, better fidelity, higher purity, less toxicity and lower loss.We choose three representative marine micro-algaes in the article. Chlorella vulgaris' ecological distribution is extensive, it is easy to plant and it grows quickly, so it is a very good meteral used to study biology technology and origin of single-cell protein. Dunaliella contains a series of available substances such as β -Carotene, protein, polysaccharides compound and microelement. In comparison with standard IAA, we detect the influence of extracts from kelp on marine micro-algaes' growth. The experimental data indicate that IAA play an important role in marine micro-algaes' growth, and 0.1~1mg/L Concentration of IAA as well as extracts from kelp can improve the level of physiological indexes such as absorbance, chlorophyll, protein and polysaccharides, we can estimate roughly the content of auxin in the meanwhile. The process lays the foundations of marine micro-algae's culture.
