Mango kernel seed is rich in polyphenols,which has great development potential.Characteristic fruit jade mountain mango kernel seed in Guangxi was used as raw material,and the extraction process,isolation and structure identification of polyphenol in which were studied.Meanwhile,the effects of antioxidant activity of the purified components from different ethanol concentration were systematically studied,which will provide a theoretical and application basis for the comprehensive development and utilization of mango nuclear products in the future.It can be drawn the following conclusion by analyzing experimental results:1.In this experiment,the ethanol was revealed to be the best extraction solventfor polyphenol from mango nuclear.The ethanol concentration,solid-liquid ratio,extraction time,extraction temperature,such as single factor experiment,then the orthogonal test and verification test would be determined.The results showed that the factors affect the extraction of polyphenols from mango kernel seed as followed:the ratio of material to liquid>ethanol concentration= extraction time>extraction temperature,optimum combination for ethanol concentration was 70%,the material liquid ratio 1:25,extraction temperature 60?,extracting time,120 min.Under these conditions,the extraction yield of polyphenol was 4.36 mg/g.The optimization of polyphenol extraction process technologu was reasonable and feasible.2.Preparation of total polyphenols:successively used macroporous resin adsorption technology,especially the AB-8 macroporous resin.The crude extracts were gained by eluting with 30%-90%ethanol after preliminary purification,separately.Methods Under the various separating condition,the polyphenols components of mango kernel seed were appraised with thin layer chromatography(TLC).The component in polyphenols was identified by TLC in three developing solution with different polarity.Two compounds with chromatography pured of 90%ethanol purified item were identified.One named K1(Rf=0.7381)was obtained,when the eluent solution was chloroform:acetone:formic acid =95:4:1.The other compound named K2(Rf=0.2857).while the eluent solution was chloroform:acetone:formic acid=93:6:1.The structure of these compounds were identified by infrared spectrum and nuclear magnetic analysis,and the results showed that the purified content from 30%and 50%were both tannic acid mainly,when that from 70%ethanol ellagic acid mainly,from 90%ethanol ricinoleic acid mainly,especially the compound K1 were determined to erucic acid andcompound K2 oleic acid glyceride.3.The results of antioxidation experiments showed that the purified polyphenols(purified by eluting with 30%-90%ethanol,K1,K2)from mango kernel seed had good scavenging effect against ·OH and DPPH· free radicals.Among them,the highest scavenging capacity of against DPPH· free radical was Vc and crude extractings from mango kernel seed,while the 70%ethanol elution components have a better ability to remove ·OH.
