Rumex.patientia L. belongs to the Polygonaceae, sorrel genus, it is a perennial herb. It has a strong ability to adapt to the environment, so rich in active ingredients, and it is a traditional Chinese medicine. In recent years, there were few reports of Rumex.patientia L. This study used Rumex.patientia L. as experimental material and it was divided into two parts of above and below the ground. Polysaccharide extracted from the two parts and then obtained the purified product by isolating and purifying the polysaccharide. The purified product was used for experiments in vitro that determined the activities of it. Significance of in vitro experiments is to provide a theoretical basis and reference basis for the development of new drugs of Rumex.patientia L. in the field of health care.The study used water extraction and ultrasonic extraction to extract polysaccharide from the two parts of Rumex.patientia L. Analyzed using Response Surface Design, the best way to extract polysaccharide obtained and can be relatively large polysaccharide content. The results showed that water extraction was better for the part on the surface and the best conditions were the temperature of 100℃,2 hours and 1:20 of solid-liquid ratio, and the maximum amount of polysaccharide extracted was 110.54mg/g. As for the subsurface part, ultrasonic extraction was the best method and the conditions included the temperature of 40℃,20 mins to extract and the solid-liquid ratio was 1:20. The maximum amount of polysaccharide extracted of the subsurface part was 130.55mg/g that was significantly more than the part on the surface.The study used water extraction and ethanol precipitation to extract crude polysaccharide from the ground and underground part of Rumex.patientia L. Using organic solvents and D101 macroporous resin elution the polysaccharide in the each part to miscellaneous, deprotein and so on. Through the DNS and TLC identification of the polysaccharide, separation and elution by DEAE-52 exchange chromatography column. Using the DNS method to determine content of the two parts from Rumex.patientia L. and the results showed that the polysaccharide in the ground part is non-reducing sugar and the content is 2.56 mg/g. The polysaccharide in the underground part is reducing sugar and the content is 2.94mg/g. According TLC analyzed the reducing sugar in underground part from Rumex.pafientia L. may contain fructose, glucose, galactose, arabia. Separation and elution the polysaccharide of two parts by DEAE-52 exchange chromatography column, the polysaccharide in the ground part eluted fractions is RLRL-h、RLPL-a、RLPL-b、RLPL-c, the content is53.33%,12.5%,24%,39.6%;the polysaccharide in underground part eluted fractions is RLRR-h、RLPR-a、RLPR-b、RLPR-c、 RLPR-d, the content is57.23%、38.35%、17.2%,25.25%、15.24%.Using the HPLC method to determine the composition of reducing sugars, and the detection is Wonda Sil C18 silicagel column, temperature: 35℃,245 nm, flow rate is 1m L/min,CH3CN: 0.05mol/LKH2PO4=2:8, 10Μl. Determine the reducing sugar contain Man,Gal,Glc and so on.Comparative study antioxidant effect, antibacterial activity and antitumor activity on polysaccharide in ground part and underground part from Rumex.pafientia L.Comparative analysis the antioxidant effect with polysaccharide and Vc. Data analysis with SPASS17.0. To study the antibacterial effect of bacteria, fungi and mycete. Observe the inhibitory effect of different concentration of polysaccharide solution samples to various strains. To study the anti-tumor activity, the polysaccharide of different concentrations was influenced on Hela cells and DK cells, and obser-ve the cell structure. The result shows that the polysaccharide in Rumex.pafientia L. has strong effect on bacteriostatic activity and Antioxidant activity Results showed that the polysaccharide in ground part scavenging effect on ?OH、O2-, DPPH stronger than the polysaccharide in underground part; the polysaccharide in underground part has stronger affect in inhibition of Bacillus subtitles, Yeast, Eumycophyta. The high concentration of polysacchariden in the underground part of inhibition to various strains were stronger than the ground. Observation of cell Structure, the polysaccha-ride in Rumex.pafientia L. can inhibit Hela cells, but has no effect on normal cells, The inhibition of the polysaccharides in underground part is stronger than the polysaccharide in ground part, and the IC50 of the polysaccharides in ground part is 54.371g/m L, the IC50 of the polysaccharides in underground part is 38.654g/m L.
