A molecular technique based on the polymerase chain reaction (PCR) provide an effective tool for the rapid identification of genetic markers known as Random Amplified Ploymorphic DNA (RAPD). It is a new molecular detective technique developed in recent years. These polymorphism allow the analysis of complex genomes without prior knowledge of DNA sequence. Moreover, RAPD is a simpler, faster, less laborious and less detective techniques. The potential use of RAPD in phylogenetic studies and population genetics has been widely documented in a large variety of organisms. The goal of this work is to study the systematic relationships among the rice grasshopper by means of RAPD analysis.In this paper, isolation of genome DNA of rice grasshopper was founded. This method can use not only in rice grasshopper but also in the insects of Odonata, Lepidoptera, Tetrigoidea. On the base of that, we compared RAPD pattern variations of 11 species, which belong to 3 genera, by using 79 different oligonucleotide primers, each with ten nucletides in length. The results show that:(1). By amplifying the same sample using the same primer, we can conclude that repetition of lsolation of genome DNA and the system of RAPD is better.(2). The primer S283 was used to amplify the genome DNA of different.rice grasshopper. The results indicate that amPlified DNA exist sexualand individua1s difference, but these difference are obviously small thanthese of species.(3). The prAners Sl30, Sl34 yield clear band in all 10 species of ricegrasshopper; the total number of bands is 45. The cluster analysissuPports the idea that O.adentata Wllemse is an independent SPecies andPSeudmp kln et Lin is an independent genus. Meanwhile, it isconsidered that O.hyla intricata (St&l) and Oj7avefemura Ma et Zhengshould be combined into one species.(4). SamPles of Omp chinensltr from six sites were also analyzed forgenetic polymorphism using RAPD. As a conclusion, we consider thatsix populations could be divided into four geograPhical races.The study results above-mentioned can suPply new basicinformation of integrated taxonomy of rice grasshopper.
