Preliminary observation of aggressive behavior of Turpan cockfighting. Polymorphism in the COMT and MAOA gene that were associated with aggressive behavior was analyzed by PCR-SSCP method in Turpan cockfighting and Xinluoman.1,Similar day-old 12 Turpan cockfightings were selected in this study. Preliminary observation of aggressive behavior of Turpan cockfighting through two tests.2,The Polymorphism of 4 Exons of COMT gene was analyzed by PCR- SSCP method.The result indicated that there was no mutation in Turpan cockfighting and Xinluoman.3,The Polymorphism of 10 Exons of MAOA gene was analyzed by PCR- SSCP method.The result indicated that only the fragments that were amplified by primer MAOA1, MAOA2, MAOA5, MAOA7 showed polymorphic. The MAOA1, MAOA5, and MAOA7 polymorphic sits was examined on Turpan cockfighting and Xinluoman, the MAOA2 polymorphic sits only was examined on the Turpan cockfighting. For MAOA1 sequencing revealed two mutations (191 Gâ†'T and 231Gâ†'A) in the genotype AB and revealed one mutation (191 Gâ†'T) in the genotype BB.For MAOA2, sequencing revealed one mutation(137 Tâ†'C) in the genotype AB.For MAOA5, sequencing revealed one mutation(207Tâ†'G)in genotype AA,AB,AC;one mutation(236 Gâ†'T)in the genotype BB,BC; one mutation(244 Tâ†'C)in genotype BB,CC,BC,AC,AB; 3 mutations(219Gâ†'A,249Gâ†'T,230Gâ†'T) in genotype CC,AC,BC; one mutation(220Gâ†'A) in genotype CC,BC;one mutation(154Câ†'T)in the genotype BB.And these mutations did not cause any amino acid change. The results of x2 test for goodness-of-fit showed that the frequencies of MAOA gene fit with Hardy-Weinberg equilibrium in two breeds(p>0.05). The results ofχ2test for independence showed that the distribution and constitution of genetypes at these polymorphic sits of MAOA gene showed significant differences between Turpan cockfighting and Xinluoman.
