The research was supported by project of "Research of the key technology ofcomprehensive utilization of sika deer blood and industrialization project"(13NK15)from Changchun science and technology bureau. The development of peptide fromblood plasma of sika deer was mainly responsible for research tasks. Three parts ofresearch work were carried out around the project research tasks, and the mainconclusions as follows:(1) Enzymatic hydrolysis preparation of peptide from blood plasma of sika deer:regarded blood plasma of sika deer as raw materials. The single protease andcompound proteases were used to hydrolysis. Neutral protease and flavorzyme weredetermined and the optimal formula ratio of enzymolysis compound was1.5:1.According to the single factor experiment, the following optimum parameters wereobtained: the hydrolysis temperature was50℃, the amount of the enzyme was5%,the pH was7, the hydrolysis time was4h, the pretreatment temperature was90℃andthe pretreatment time was15min. According to the above experiments, the responsesurface methodology was used to obtain the optimum hydrolysis parameters: thehydrolysis temperature was51.2℃, the amount of the enzyme was5.82%and pH was7.16, the degree of hydrolysis was18.41%. Comparing blood plasma of sika deerantioxidant activities before and after enzymolysis, the DPPH radical scavengingactivity, ABTS radical scavenging activity and reducing power of blood plasmahydrolysates from sika deer were all higher than blood plasma of sika deer.(2) Preparation of powder of blood plasma peptide from sika deer by spraydrying technique: Study on the effects of the ratio between hydrolysate solids anddrying aids on spray drying, determined the optimum ratio was6:4. According to the single factor experiment, the optimum parameters were determined as follows: thematerial concentration was30%, the inlet air temperature was160℃, theapproachingspeed was6ml/min and the compressed air flow was601L/h. On the basis of singlefactor experiment, setting powder rate and moisture content as index by L9(34)orthogonal experiment method. The three factors material concentration, inlet airtemperature, approaching speed were optimized. The optimum drying parameterswere as follows: the material concentration was30%, the inlet air temperature was180℃and theapproaching speed was9ml/min. Under the optimum dryingparameters, the collection of powder rate was47.52%, moisture content was3.26%.(3) Granules of the development of blood plasma peptide from sika deer:according to the single factor experiment, the following parameters were obtained: theamount of adding sweetener stevia sugar was0.5%, the amount of the lactose was20%, the wetting agent was ethanol. Study on the effects of alcohol content, dryingtemperature and drying time on granulating efficiency, the optimum parameters weredetermined as follows: the concentration of the ethanol was80%, the dryingtemperature was60℃and the drying time was30min. According to the aboveexperiments, the response surface methodology was used to obtain the optimumgranulating process parameters: the concentration of ethanol was83%, the dryingtemperature was60℃and the drying time was32min. Under the optimum dryingparameters, the granulation forming rate was88.92%, sensory evaluation, physicalchemical index and microbiological index were determined.
