Deer blood is the blood of cervidae animal sika deer or red deer, is a kind ofprecious traditional Chinese medicine, which had been recorded in Chinese herbalmedicine of past dynasties, and folk also widely circulated. Deer blood has anti-aging,enhance immunity, blood tonic, fatigue resistance and nourishing liver, and otherfunctions. As people pay more attention to the health food and medicine, makemedical value of deer blood has been attention at home and abroad. The application ofliving blood sampling technique further enriching the deer blood resources, providethe conditions for the development of deer blood products. But deer blood productsare mainly primary processing, product level is low, less intensive processingproducts, the development is still inadequate. In this paper, by centrifugal separationtechnology to separate the deer blood plasma and blood cells. According to theirnutritional characteristics applying different processing methods. Optimized the deerplasma spray drying processd and deer hemoglobin enzymatic process, studied theantioxidant activity of enzymolysis products and its microencapsulation, measured theindicators of microcapsule product.The main research contents and results are as follows:1. In the separate experiment, to plasma yield, hemoglobin concentration andprotein concentration in plasma for the index, single factor analysis of variance wasadopted to determine the effects of the centrifugal rotational speed and the centrifugaltime on the separation efficiency, Results show that: the centrifugal rotationalspeed at4000r/min, centrifugal time was10min, the deer plasma had yield large,low hemoglobin concentration, high total protein concentration, good centrifugalseparation.2. First by single factor experiment, research into the inlet temperature, feedrate and air velocity on the effects of the spray drying, With the moisture contentand powder rate and nitrogen solubility index (NSI) as the index, orthogonalexperiment to optimize the process conditions.The results as follows: inlettemperature of160Co, feed rate of300mL/h, air flow36m3/min.3. Deer blood cells hemolysised by ultrasonic, prepared deer hemoglobin solution. From the neutrase, alcalase, papain and pancreatin chosed the neutrase as theoptimum enzyme. On the basis of single factor test, optimum conditions to achievethe maximum antioxidant ability were determined by response surface methodology:substrate concentration9%, hydrolysis time4hours, hydrolysis temperature56Co,enzyme concentration8255U/g, and pH10.2.Under these conditions, reducibilityreached0.811, showed good antioxidant activity.4. The research onbantioxidantion of peptides hydrolyzed from deer hemoglobinby enzyme. With the deer hemoglobin antioxidant peptide concentration at15mg/mL,the reducibility reached0.778, when concentration was10mg/mL, theclearance rate to DPPH free radical was91.75%, when concentration was30mg/mL,the hydroxyl radicals on the removal rate was95.4%.5. On the basis of single factor experiment, seven independent variables of spraydrying process were studied on microencapsulation, including the porous starchconsumption, the proportion of wall material (Arabic gum: maltodextrin), ratio ofcore and wall material, solids content, inlet temperature, feed rate and air velocity.Using four factors quadratic general rotational combination digestion conditionsoptimization, the optimum technological conditions as follows:the the porous starchconsumption of2:1, the proportion of wall material is1:1, ratio of core and wallmaterial of1:1.0, solids content22%, inlet temperature of140oC, the optimal feedrate is280mL/h and the air flow into the24m3/h. The microencapsulation efficiencyof products obtained under these conditions was95.93%by validation test.6. Determinated the moisture, the bulk denstity, the solubility, the caesura angle,and other physical properties of deer hemoglobin antioxidant peptide microcapsuleproducts, the results showed that the microcapsules was3.74%moisture content,suitable for long-term storage, bulk density is0.39g/cm3, angle of repose is38o, thesolubility is74.65%. And the SEM showed that the particle size of deer hemoglobinantioxidant peptide microcapsule product was given priority to with8~25μm, is closeto spherical particles with smooth surface and compact, no cracks. Microcapsuleproducts with certain slow-release and it showed that in the75%RHcondition,microcapsule products stored stably.
