Counter-Current Chromatography is a liquid-liquid partition chromatography. It needs no stationary phase supporter, so it overcomes the irreversible adsorption in conventional partition method. It has a high recovery rate, a wide range of application, an easy instrument operation, and a large partition scale. As a result, it is applied widely in the separation and purification of active substance in natural products.Now, research workers have established several math models of CCC, such as cell model, countercurrent distribution model, Continuous-Stirred Tank Reactors model, probabilistic model. Based on plate theory and Van’t Hoff equation, a new temperature dependence plate model has been set up in this paper and obtained result below:Using Matlab to do computer programming, the modeling of the partition action of CCC can be realized. New program was applied in prediction of experimental result, and the result is good. Several other experiment parameters were introduced to adjust the math modeling result, and it decreased the error in Matlab modeling prediction. By studying the number of plate and the peak shape of CCC, the plate efficiency parameter was introduced to the plate model, and the errors of predicted peak shapes were greatly reduced.The partition coefficient, KD, of some flavonoids, in a two-phase solvent system composed of ethyl acetate-methanol-water (50/1/50, v/v), was measured at six specific temperatures ranging from20to45℃by a slow stirring approach. The analyzing results are helpful for us to choose the suitable operation conditions for high throughput products.When temperature was taken into consideration, the error of retention time was significantly reduced. For one peak, the retention time was reduced from9.22%to nearly zero, and for another peak, the retention time error was reduced from5.46%to nearly zero.Using Matlab software as means of realization, this paper realized CCC’s CSTR model by not complicated math modeling method. This paper found the relationship between partition parameters and math modeling, and it gave new idea and research method to theoretical research of CCC.
