Mikania micrantha is one of the worst invasive alien species.It has caused much more damage to ecosystems in our country.In order to prevent and control Mikania micrantha,it is necessary to separate and analyze the active components in the natural plant.Flavonoids are claimed to possess various pharmacology effects.They are major functional components of many herbs for medical use.So how to effectively separate these virtual components becomes especially crucial.As a support-free liquid-liquid chromatographic technique where the mobile and stationary phase are composed of two immiscible solvents or solutions,high speed countercurrent chromatography(HSCCC)has many advantages,such as economical,high capacity,and high purity.It has been a promising method for separating and purifying flavonoids from natural products.In this study,flavonoids were separated from Mikania micrantha by HSCCC.The main results were as follow:1.Established a method for the separation and purification of four flavonoids from Mikania micrantha by using macroporous resin combined with HSCCC.Macroporous resin was AB-8 and the eluent was 50%(v/v)ethanol.A two solvent system composed of n-butanol/acetic acid/water(4:1:5,v/v)was used in HSCCC.Quercetin-3-O-rutinoside,kaempferol-3-O-rutinoside,luteoloside and astragalin with the purity of 90.12%,98.55%,98.33%and 99.23%,respectively,were successfully separated from Mikania micrantha by HSCCC in one run.This method is simple and efficient and was used in the following separation of flavonoids from invasive plants.2.Designed an on-line magnetic nanoparticles with countercurrent chromatography device.Effects of flavonoid adsorption and desorption of several magnetic nanoparticles were studied.On-line magnetic nanoparticles with HSCCC device was used for separation of flavonoids from Mikania micratha.The method could be used in further separation of flavonoids from other natural products.
