This paper has reviewed the recent application of high speed countercurrent chromatography in the separation of Chinese traditional herbs. It has also obtained the rules of selecting the solvent systems of high speed countercurrent chromatography and found some simple but prompt methods to choose solvent system. Finally it used these methods to get some solvent systems and separated some alkaloids from Coptis chinensis Franch phellodendron chinense schneid Tinospora capillipes Gagnep Mabonia bealei(Fort)Carr Corydalis turtshaninovii Bess. f. yanhusuo Y.H. Chow et C.C. Hsu Nandina domestica Thumb and Ephedra sinica Stapf.The optimum range of the partition coefficient values (K) for solvent system of high speed countercurrent chromatography should be between 0.5-2. And there has two ways to calculate it. One is simply to detect the absorbance of light or the quality of each part of solvent system when thesample is a pure material; The other is to calculate by HPLC when the sample is an impure one, we can counter the peak area of each fraction of solvent system which is measured by HPLC. The ratio of upper phase to lower phase is the partition coefficient K.The separation condition is as below: the flow-rate of mobile phase is 2 ml/min; the speed is 850 r/min; the detection wavelength is 254 nm. Palmatine berberine epiberberine and coptisine were separated from Coptis chinensis Franch Palmatine berberine and 4-Hydroxy-3-methoxycinnamic acid were separated from phellodendron chinense schneid;Palmatine from Tinospora capillipes Gagnep: Palmatien and berberine from Mabonia bealei(Fort)Carr Dehydrocorydaline from Corydalis turtshaninovii Bess. f. yanhusuo Y.H. Chow et C.C. Hsu; Berberine from Nandina domestica Thumb; Ephedrine from Ephedra sinica Stapf.It also talked about the separation factor of hsccc which is include temperature the flow-rate of mobile phase and the speed in this paper. You can also see the future of high speed countercurrent chromatography from the paper.
