Reserches On Chemical Composition、Analysis And Extraction Technology Of Alkaloids From Cyclea Hypoglauca(Schauer) Diels

In this study,the root of Cyclea hypoglauca（Schauer）Diels was used as the raw material to determine the content and extraction of alkaloids.The chemical components in the extract were studied.1.Analysis the chemical compositions of the extract from Cyclea hypoglauca（Schauer）DielsHPLC-Q-Exactive Focus-MS/MS technology was used to analyze the components of the extracts from Cyclea hypoglauca（Schauer）Diels.139compounds were inferred and identified,including 29 alkaloids,3 sterols,2sterones,and 10 polysaccharides,2 lignins,31 organic acids,5 phenols,1flavonoid,5 amides,14 fatty acids,4 amino acids and other compounds.2.A method was established to detemine the content of alkaloidsThrough a serier of conditions exploration,HPLC was used with Wetch Xtimate C₁₈ column（4.6x250mm,5μm）.to detect the content of alkaloids.The mobile phase was acetonitrile（A）and 0.5%phosphoric acid aqueous solution（B）,and the gradient elution was 0～10min,5%→10%A;10～60min,10%→18%A;60～70min,18%→25%A;70～85min,25%→30%A;85～100min,30%A,detection wavelength:282nm,flow rate was 0.5 m L/min;Column temperature was kept at30℃.Through methodological verification,the method is accurate,reliable,sensitive,quickand reproducible.This method can provide experimental basis for the quality evaluation and control of the alkaloids of the extraction from Cyclea hypoglauca（Schauer）Diels.3.Research on the extract process of alkaloidsUltrasonic and microwave-assisted extract was used to study the optimal extract process of the alkaloids in Cyclea hypoglauca（Schauer）Diels.Firstly,the four factors with greater influence were screened out through single factor experiment:ethanol concentration,material-to-liquid ratio,p H and extract times.Afterwards,through orthogonal optimization experiments,the optimal extract conditions for alkaloids were:Cyclea hypoglauca（Schauer）Diels powder;liquid to raw material ratio,1:18;p H of the extracting solution was adjusted to 2;ethanol concentration,60%,extraction time,30min,ultrasonic power was 200W;microwave power was 400W;extract temperature was 80°C;extract time was 1h;extract was performed four times.The highest extract rate of alkaloids from Cyclea hypoglauca（Schauer）Diels was 2.33%.
