Phellinus has anti-tumor, anti-cancer, anti-liver fibrosis and anti-lipid peroxidation activities.It is widely recognized as the best anti-cancer medicinal fungus. Polysaccharide is the main physiologically active Phellinus active ingredients. Phellinus and polysaccharide products are widely used in pharmaceutical, food, clinical treatment and cosmetics fields. Today Phellinus polysaccharide is mainly from the fruiting bodies and submerged mycelium culture.As the lack of resources of wild Phellinus, artificial cultivation technology is not yet mature, liquid fermentation has high cost, sophisticated equipment, heavy pollution and other shortcomings, this study uses solid-state fermentation cultured Phellinus mycelium, and uses the single factor and orthogonal Optimization optimized the liquid-culture medium and solid-culture medium respectively.Finalizing the liquid medium is soy meal 3%, soluble starch 3%, MgSO 4·7H 2O 0.5 g/L, KH 2 PO 41 g/L, pH 6.0, mycelium conversion rate can reach 40%, the best time to take seed liquid is 9-12 days. Optimizing the factors, ultimately determine the solid state fermentation substrate is A, filling capacity is 30 g, CaCO 3 addition rate is 10%, the ratio is 5/4, then the sugar content is relatively the highest mycelium.Intracellular polysaccharide extracted by hot water extraction. Respectively, optimizing the extraction processes of wild Phellinus polysaccharide and solid-culture Phellinus polysaccharide, and finalizing the wild Polysaccharide extraction process:the extraction temperature is 90℃, solid-liquid ratio is 1/50, the extraction time is 4 h. The extraction rate of wild Phellinus polysaccharide is about 0.028%; solid-culture Phellinus polysaccharide extraction process:the extraction temperature is 90℃, solid-liquid ratio is 1/50, the extraction time is 3 h.The extraction rate of the solid-culture Phellinus polysaccharide is about 0.25%. The polysaccharide content of solid-culture Phellinus is about 10 times the wild Phellinus.We using Sephadex G-100 gel chromatography to determine that the best ethanol concentration is 30%. Detecting polysaccharide molecular weight distribution by gel permeation chromatography (GPC), we found that polysaccharides from solid-state fermentation, PI固, Mw is 2.072×104, and polysaccharides from fruiting bodies, PI子, Mw is 1.785×104. The infrared spectras of two polysaccharides show that they both contain absorption peaks of saccharide, mainly are the O-H stretching vibration and C-H stretching vibration, as well as weak C=C stretching vibration. Analyzing polysaccharide structures by infrared spectroscopy, monosaccharide component of PI固is glucose, and monosaccharide component of PI子is fructose.Using MTT assay efficacy, we found that 30% ethanol precipitation of the purified wild Phellinus polysaccharide and Phellinus polysaccharide fermentation both have directly weak toxic effect on the Huh7 cells.
