Sanguisorba officinalis L is the Rosaceae Sanguisorba perennial herb, Widely distributed in our country, Especially in eastern Inner Mongolia, Heilongjiang, jilin, liaoning and other regions, The wildlife reserves accounted for more than 70% of the country. In the garden burnet polysaccharide content is higher, The pharmacological studies have demonstrated that Sanguisorba polysaccharide is a kind of new alpha glycosidase inhibitor, can have fall blood sugar blood fat, increase immunity of physiological functions. Has high research value and broad prospects for development. Sanguisorba as the research object, this paper on its polysaccharide extraction, isolation, purification and performance study, Aimed at Sanguisorba polysaccharide of comprehensive utilization of product development and provide a reference basis. The main results were as follows:1. Test respectively use the ultrasonic extraction method and the composite enzyme extraction YuZhong extracting polysaccharide from the ground, Results show that the composite enzyme extraction method is superior to the ultrasonic extraction method, The method extraction rate is higher, the optimal extraction conditions for cellulose is 1.5%, Papain is 1.5%, temperature is 30 ℃, enzymolysis time 1 h, pH 5.0, plead liquid ratio 35:1, Under the condition of the polysaccharide extraction yield of 13%. Using phenol- sulfuric acid method,it’s determination of polysaccharides in crude polysaccharides content to its purity is 31.15%.2. It’s use macroporous resin chromatography to remove Sanguisorba polysaccharide extract the polyphenols and determine the optimal purification conditions as follows: First,Choose HB- 1600 as the best adsorption resin, In Sanguisorba polysaccharide on sample concentration of 0.333 mg/mL, column on the flow rate of 1 BV/h, then, elution velocity conditions for 1 BV/h, it can make the garden burnet polysaccharide to 82.88%, and the removal rate of polyphenols in Sanguisorba polysaccharide purity from 31.15% to 76.50%.3. It can test respectively by enzymatic hydrolysis and Sevag method to remove Sanguisorba polysaccharide protein containing impurities, Through test analysis to determine choice papain enzyme method is more appropriate, the optimum adding amount of 0.2 mL(800μ), the best enzyme solution temperature of 60 ℃, then, the best time to enzyme solution for 2h; The sevag method in addition to protein with three times advisable, the vibration wave time best for 35 min. Finally it through the GPC detection, in addition to protein before and after the samples detection by Sevag method, the enzymatic hydrolysis in combination with deproteinization effect is better and protein clearance rate reached 42.11%, the polysaccharide retention rate is 81.30%; the Sanguisorba polysaccharide purity up to 85.6%.4. By the research on the stability of Sanguisorba polysaccharide, the results show that Sanguisorba polysaccharide in weak alkaline, metal ion, REDOX concentration is low, and avoid light low temperature(< 60 ℃) under the condition of more stable; Experimental studies on the oxidation resistance of Sanguisorba polysaccharides, the results show that the ability to remove hydroxyl free radical is 28%, remove the ability of DPPH free radical is 40.4%, and slight is weaker than Vc antioxidant capacity; For its bacteriostatic effect was analyzed, and the results show that the lowest Sanguisorba polysaccharide on e. coli bacteriostatic concentration(MIC) of 10 mg/mL, bacillus subtilis, minimum bacteriostatic concentration(MIC) of 10mg/mL, the staphylococcus aureus minimum bacteriostatic concentration(MIC) of 10 mg/mL.
