Tara?Caesalpinia spinosa Kuntze?is a rare species which comes from South America as a substitute for the preparation of gallnut tannic acid series products in China.It is the seed of Tara bean,with a thick layer of endosperm,and its main ingredients are manifolds of sugar.According to the existing related research,it is found that Tara mannan polysaccharide is a kind of galactoglucomannan.This experiment is mainly to study on the extract of Tara seed polysaccharide,and the removal of protein,optimize the decolorization process,the chemical structure,analyzes the structural characteristics of antioxidant.These provide a valuable theoretical reference to the comprehensive application in food medicine,chemistry and other industries about Tara polysaccharide.1.Three methods of hot water extraction,cold Waterlogged extraction,and ultrasonic assisted extraction were used to extract Tara polysaccharide.Through single factor experiments,it is found that ultrasonic extraction was the best extraction method.Therefore,on the basis of single factor experiment,the experimental condition range of higher extraction rate was chosen,and the response surface optimization experiment was carried out to optimize the experimental parameters,the final optimum extraction conditions the ultrasonic power of 363w,ultrasonic temperature 56,ultrasonic time 23 min,material to liquid ratio of 1:17?g/ml?,the maximum extraction rate of Tara crude polysaccharide was 25.56%and the theory value of 25.71%are small.The SEM results also showed that the extraction is sufficient.2.Three methods of TCA,Sevag,papaya protein enzymatic were used to remove protein Tara form coarse polysaccharide.According to single factor experiment,it had a better effect that use Papaya protein enzymatic protein.The parameters were optimized through orthogonal experiments,and the final optimum conditions for eliminating protein was that enzyme dosage is 0.15ml,enzymatic hydrolysis time is 90min,hydrolysis temperature was 60?,enzymolysis is pH 6.0,protein removal rate is 95.19%,the retention rate of polysaccharide is 75.02%,and the yield of polysaccharide is 19.17%.3.Granular activated carbon,active carbon powder,H2O2 were applied on the extraction of Tara crude polysaccharide decolorization experiment.It found that granular activated carbon has the best Decolorization effect.The experimental parameters were optimized through orthogonal experiment,which concluded that the4.optimal decolorization conditions for activated carbon particles is 0.6g,decolorization time is 60min decolorization,temperature is 45?,3 decolorization times,the decolorization rate is 50.55%,the retention rate of polysaccharide is 92.92%,and the final yield of 17.81%.Polysaccharide.5.IR data showed that Tara polysaccharide has a beta type glycosidic bond and the general characteristics of polysaccharides.GPC pattern showed,which Tara polysaccharide is different from galactomannan and more similar to Grape mannan,which the Number-average Molecular Weight in?36?*104.XRD analysis showed that Tara polysaccharide has no obvious strong absorption peaks at Ze 10400 range,but there are only a few small peaks.It is indicated that the Tara polysaccharide formed single amorphous crystal under the condition.It is also showed the same results by X-ray diffraction that Tara polysaccharide is amorphous by DSC,TG map.6.Through the Tara polysaccharide oxidation experiment under different purity and concentration,it is found that after purification of IC50 was lower than that before purification With VC as positive control of crude and antioxidation of Tara polysaccharide.It is indicated that the higher purity of Tarado sugar has better oxidation resistance,and the higher concentration of ABTS radical,DPPH radical and hydroxyl radical scavenging,the ability is stronger.Otherwise,the highest sugar Tarado radical scavenging rate on ABTS can reach96.98%after purification.The highest adical scavenging rate on DPPH can reach 92.99%.The highest of hydroxyl radical scavenging rate can reach 99.79%.This suggested that Tara polysaccharide has a good oxidation resistance,and can be used as an antioxidant.
