In this study, we will increase the number of rounds of amplification rounds, and the effect of amplification specificity by the combination of nano material quantum dots and multiple wheel gene amplification. The quantum dot in the high fidelity Pfu polymerase catalyzed by the PCR system can produce a significant thermal effect. In this project, we first select 3 kinds sources of template and high GC content of the primers based on quantum dots based on multiple rounds of gene amplification, to explore the factors affecting the amplification specificity. To marine biofouling barnacle M8 template multiple rounds of amplification experiment, using quantum dots in the experiment the amplification rounds from 3 rounds of amplification increased to eight rounds in, through compared to the same amplification conditions conventional multi-round gene amplification and nano gold multiple rounds of gene amplification method based amplification effect, further validation of the quantum dots for several rounds of gene amplification specificity of raise has a significant effect. Through comparison of quantum dots of different templates and template of low concentration multiple rounds of gene amplification effect, and different surface modification of quantum dots on several rounds of gene amplification effect influence, quantum dots showed stable and excellent synergistic effect, on the basis of the amplified products were sequenced and the results are compared, showing the quantum dots of several rounds of gene amplification with high fidelity, which can have important significance as a practical application.Quantum dots in multiple rounds of gene, catalyzed by the PFU polymerase amplification can produce obvious thermal starting effect, and in ordinary Taq polymerase catalyzed amplification system without significant difference. Therefore, between the quantum dots and PFU polymerase interaction as the main mechanism of the research object. Preliminary detection of PCR system in each group were able to produce certain effect and quantum dots by agarose gel electrophoresis, but between quantum dots and PFU polymerase has the greatest impact. After the UV visible light spectrum method and Fourier transform infrared spectroscopy in deep research and analysis confirmed the interaction between quantum dots and PFU polymerase.
