The identification of mushrooms and their spores is of great significance for the study of fungi.Pollen identification and classification are meaningful to biology and consumers.In this dissertation,Fourier Transform Infrared Spectroscopy?FTIR?and Surface Enhanced Raman Spectroscopy?SERS?were used to identify Tricholoma matsutake and Agaricus blazei,Cedar?Pinus massoniana Lamb?and bee pollen,the spores of three kinds of Boletus,Amanita pantherina,Lactarius hatsudake Tanaka and Russula virescens.The main conclusions are as follows:1.The FTIR spectra of Tricholoma matsutake and Agaricus blazei were mainly composed of the absorption of protein and Polysaccharide.The intensity and position of the peaks were different in the range of 1650-750 cm-1 in SD-IR spectra.In the range of 1666-1388cm-1 and 1120-918cm-1,the number and intensity of the auto- matic peaks in the 2D-IR spectra of Matsutake and Agaricus blazei were different.Curve fitting results in the range of 1186-1000 cm-1 showed that the area ratios of the sub-peaks were different.The surface-enhanced Raman spectra of Agaricus blazei showed Raman signals at positions 1655,1574,1444,1340,1296,1223,1134,532,and 474 cm-1.The intensity,number and location of Raman signals of Tricholoma matsutake and Agaricus blazei were different.2.In the range of 4000-400 cm-1,the average FT-IR spectra of Pinus massoniana Lamb and eight bee pollens were similar.The SD-IR spectrum of Pinus massoni-ana Lamb exhibited strong peaks at 1707 cm-1,while camellia bee pollen,lotus bee pollen,rose bee pollen,buckwheat bee pollen,schisandra bee pollen,canola bee pollen,motherwort bee pollen and corn bee pollen showed strong peaks at 1747,1738,1745,1736,1741,1741,1743 and 1740cm-1.The 2D-IR spectra of cedar and bee pollens show the number and intensity of automatic peaks were different.The results of curve fitting show that the percentage of peak area of bee pollen and cedar pollen were different.The morphology of cedar pollen was significantly different from that of eight bee pollens under scanning electron microscopy,but there was no significant difference among eight bee pollens.The surface-enhanced Raman spectra of cedar pollen and bee pollens were significantly different,and the intensity,number and position of Raman signals were different.3.The microscopic morphology of three Boletus spores is similar.The size of the spores of Amanita pantherina,Russula virescens and Lactarius hatsudake Tanakawere all approximately circular in size between 5.5 and 6.4 um,but their surfacetexture were slightly different.The location and intensity of surface enhanced Raman peaks of the spores were different.The results show that the two-dimensional correlation infrared spectroscopy combined with surface-enhanced Raman spectroscopy could identify Tricholoma matsutake,distinguish pollen and mushroom spores.
