Abstract/Summary:
LncRNAs are ubiquitous RNA moleculer playing significant function in the process of development. It can’t translate to protein, and usually regulate the expression of gene by transcription. With the progress of high-throughput sequencing, many LncRNAs have been found, but the function of many novel LncRNAs are stil not clear.Using RNA-seq data, a novel LncRNA locating in mouse chromosome 2 was predicted. It specifically express in brain of mouse embryo.After verifying the expression of LncRNA –Scrt2 in mouse E15.5 brain, This research use RACE to get full lengh of transcription that is 1582 bp. Then, this research further proved that LncRNA-Scrt2 is a LncRNA. First, by ORF finder, this research found all ORF of transcription is less than 300 bp. That is to say LncRNA –Scrt2 can’t translate to protein.Next,by cell fish, this research detected that LncRNA-Scrt2 express in cell nucleus.It is also can prove that LncRNA-Scrt2 is a LncRNA.This research analyzed the expression patern of LncRNA –Scrt2 in mouse embryo. By QRT-PCR, this research detected LncRNA- Scrt2 dynamically expressing in brain of mouse embryo from E12.5 to E18.5, and there are peak in E15.5. The results of in situ hybridization showed that LncRNA-Scrt2 was specifically expressed in the brain tissues during E10.5 days, E12.5 days and E15.5 days, and the expression of the brain also changed with the development of the brain. To further observe the expression of E15.5 in the brain, we ues fluorescence in situ hybridization and found that the expression of LncRNA-Scrt2 was mainly concentrated in the ventricles of the brain and the sub regions of the brain, and the expression of the inner layer to the outer layer decreased in turn.In summary, this study found a novel LncRNA-Scrt2 related to mouse embryonic brain development.And then we get the full length of transcription. the transcript sequence analysis proved that it is a LncRNA. The expression profile of mouse embryonic development stage showed that the specific expression in the mouse embryonic brain tissue. This study deepens the understanding of LncRNA in the brain, and provides a good experimental basis for further functional study of LncRNA-Scrt2.
